“…Cellulases produced by certain microorganisms can play a key role in the degradation of cellulose present in stone cells (Ajeje et al, 2021; Ren et al, 2019). These cellulases include exoglucanase, endoglucanase, and β‐glucosidase (Talebi et al, 2022). Studies have shown that clear zone test on CMC medium with iodine and the filter paper degradation test are the most common methods to test the production of cellulase by microbial strains (Kurnia Ila & Astuti Feb, 2021).…”

“…A 2.0 mL sample of the 1-month-subcultured microflora was centrifuged at 14,000× g and 4 • C for 10 min. The resultant supernatant, termed enzyme solution, was analyzed for the presence of cellulase, chitinase, and pectinase using the modified dinitro salicylic acid (DNS) method [21]. In brief, 200 µL of the enzyme solution and 800 µL of 50 mM citrate buffer (pH 4.8) containing 12.5 mg/mL of carboxymethyl cellulose, chitin from shrimp shells, or citrus pectin, as a substrate for cellulase, chitinase, or pectinase, respectively, were mixed in a microtube and incubated for 1 h at 50 • C. The tubes were then centrifuged at 14,000× g and 4 • C for 10 min.…”
