An Ultrasensitive Colorimetric Strategy for Detection of Cadmium Based on the Peroxidase-like Activity of G-Quadruplex-Cd(II) Specific Aptamer

Abstract: We rationally designed an ultrasensitive and label-free sensing platform for determination of cadmium (Cd). The sensing platform contains G-quadruplex-Cd(II) specific aptamer (GCDSA) constructed by incorporating G-rich sequence at the end of 5′ and the critical domain of the Cd-4 aptamer. GCDSA designed act as both a special recognition sequence for Cd 2+ and a signal DNAzyme. In absence of Cd 2+ , GCDSA may mainly exist in a random coil sequence. Upon addition of Cd 2+ , GCDSA could probably be induced to fol… Show more

“…This method avoids using labeled oligonucleotides, makes the experimental procedure simple and convenient, allows for direct quantitative sample analysis, and has an excellent dynamic range. It displays two segments with a broad linear range, allowing Cd 2+ to be detected at concentrations as low as 0.15 nM [ 71 ].…”

“…For example, Tao et al [3] used gold nanoparticle modified MoS 2 nanocomposites as an enzyme mimic to enhanced peroxidase-like activity and improved the sensibility of colorimetric Cd 2+ detection. Zhou et al [10] designed a G-rich sequence for the aptamer to bind with Cd 2+ and form a G-quadruplex structure as a peroxidase-like DNAzyme to enhance the catalytic activity of heme in order to provide a fluorescent signal for cadmium detection. Zhu et al [11] decorated Carboxyfluorescein(FAM)-labeled 3 end and poly-G structure 5 end to the aptamer and its conformation changed from a random coil sequence to a stem-loop structure when it met Cd 2+ and this resulted in the fluorescence of FAM change which linked with Cd 2+ concentration.…”
