Anagliptin, a dipeptidyl peptidase‐4 inhibitor, improved bladder function and hemodynamics in rats with bilateral internal iliac artery ligation

Abstract: Aims To investigate the effect of anagliptin (Ana), a dipeptidyl peptidase‐4 (DPP‐4) inhibitor, on acute ischemia‐induced bladder dysfunction in rats. Methods Eight‐week‐old female Wistar‐ST rats were randomly assigned into four groups: (a) sham; (b) ligation (Lig); (c) Lig + Ana; and (d) Lig + Liraglutide (a glucagon‐like peptide‐1 [GLP‐1] receptor agonist; Lira). Rats in the Lig, Lig + Ana, and Lig + Lira groups underwent ligature of the bilateral internal iliac arteries. Ana was orally administered mixed wi… Show more

“…Awake cystometry was performed as previously described. [13][14][15] Briefly, under isoflurane anesthesia, the rats' abdomen were incised along the midline to expose the bladders. A polyethylene catheter (PE-50) was inserted into the bladder dome and secured with a 5-0 suture.…”

“…After the neurological evaluation, all rats received cystometry according to methods described previously [21,22,23]. With inhalation anesthesia (4% isoflurane for induction and 1.5%–2.0% isoflurane for maintenance), the abdomens of the rats were opened through a midline incision, and the bladders were exposed.…”

“…Chronic hypoperfusion due to atherosclerosis of pelvic arteries is considered as a cause of bladder dysfunction (Michel et al 2015a ; Thurmond et al 2016 ). One study explored effects of a 4-week treatment with the dipeptidyl-peptidase 4 inhibitor anagliptin and the glucagon-like peptide 1 (GLP-1) receptor agonist liraglutide in a preventive setting in a rat model of acute bilateral internal iliac artery ligation (Hotta et al 2020 ). Neither the ligation nor the treatments affected glucose levels or bladder weight.…”
