Analgesic and Antioxidant Activity of Mangiferin and Its Derivatives: the Structure Activity Relationship

Abstract: Mangiferin, 2-b b-D-glucopyranosyl-1,3,6,7-tetrahydroxy-9H-xanthen-9-one, obtained directly from methanolic extracts of Bombax ceiba leaves in substantial amounts demonstrated strong antioxidant activity (EC 50 5.8؎0.96 m mg/ml or 13.74 m mM) using DPPH assay comparable to rutin, commonly used as antioxidant for medical purposes. The acetyl and cinnamoyl derivatives were found to be less active than mangiferin whereas, methyl and 3,6,7-trimethylether tetraacetate derivatives were inactive implying that for ant… Show more

“…The antioxidant activity is traceable already in the healthy condition, but it is strikingly amplified in the inflammatory and infectious, including diabetes, states [7,8]. In our studies, mangiferin decreased diabetic-enhanced lipid peroxidation products -thiobarbituric acid reactive substances (TBARS) formed as degradation products of fats -down to the healthy level.…”

“…Moreover, mangiferin showed hepatoprotective activity against carbon tetrachloride induced liver injury further supporting the free radical scavenging property in the in vivo system. Another molecules acetyl and cinnamoyl derivatives were found to be less active than mangiferin whereas, methyl and 3,6,7-trimethylether tetraacetate derivatives were inactive (Dar et al, 2005). Vieira et al (2009) also evaluated the antioxidant activity of methanolic extract of Bombax ceiba using (i) ability to scavenge DPPH and hydroxyl free radicals; (ii) action against lipid peroxidation and (iii) effect on myeloperoxidase activity.…”

“…It possesses several pharmacological activities like antioxidant (Rodriguez et al, 2006), anti-inflammatory (Saha et al, 2016), anti-viral (Zheng and Lu, 1990), analgesic (Dar et al, 2005), immunomodulatory (Leiro et al, 2004;Sarkar et al, 2004), antidiabetic (Ojewole, 2005), besides other's. Amongst all these, the ability of Mgf to act as a potent antioxidant by scavenging free radicals is known to be of immense importance for preventing the potential DNA damage and for inhibiting the growth of cancer cells by inducing cellular apoptosis (Minglin et al, 2016;Takeda et al, 2016).…”
