The analytical capabilities of liquid chromatography tandem mass spectrometry for sensitive and highly selective determination of target compounds in complex biological samples makes it well suited for high-throughput analysis. We report the fast separation of six benzodiazepines isolated from human urine via selected reaction monitoring liquid chromatography/mass spectrometry using short dwell times to accommodate fast-eluting chromatographic peaks. The analytes were extracted from human urine samples along with their deuterium-labeled internal standards by a simple liquid-liquid extraction in 96-well plates. Using four autosamplers coupled to one chromatographic column and one tandem mass spectrometer operated in the turbo ion spray mode with positive ion detection, 1152 samples (12 96-well plates) were analyzed in less than 12 h. Through an electronic switching box designed and constructed in-house, the autosamplers were synchronized with the mass spectrometer so that injections were made as soon as the mass spectrometer was ready to collect data. Each run required 30 s to complete with another 7-8 s for the data system to load the next data file to be collected. Chromatographic integrity and ion current response remained relatively constant for the duration of the analyses. The results show acceptable precision and accuracy and demonstrate the feasibility of using fast separations with tandem mass spectrometry for high-throughout analysis of biological samples containing multiple analytes.

Although alkaloids are naturally occurring bases, additional generalizations are difficult because they include a wide range of structurally dissimilar compounds. They vary greatly in their chemical and physical properties as well as in their distribution in nature. In some cases certain alkaloids are associated with only a single species of plant, whereas others are more widely distributed between biological groupings of plants, and yet others are found in a wide range of unrelated plants. Likewise, the concentration of a particular compound may be highly localized within a given region of one plant and found predominantly in a different region of another plant. In some cases the levels of an alkaloid may be relatively high and its isolation as a natural product may be economically feasible, whereas in other cases the levels may be extremely low and less commercially desirable. The medicinal use of alkaloids in the form of crude plant extracts has been known for several thousand years and today there are hundreds that have been isolated and characterized. However, very few of them are accepted therapeutically and many of these fit into the broad categories of cinchona, ergot, opium, rauwolfia, tropane, vinca and xanthine alkaloids.

The content and dissolution rate of theophylline, diprophylline and proxyphylline from a sustained release formulation were determined by UV in situ densitometry. After separation the chromatographic zones corresponding to the spots of theophylline, diprophylline and proxyphylline on the high performance thin layer chromatographic plates were scanned in reflectance/absorbance mode at 275 nm. Quantification was performed with a second degree polynomial function over the range 40-200 ng for theophylline and 60-300 ng for diprophylline and proxyphylline. Percentages of dissolved theophylline, diprophylline and proxyphylline were monitored over 1, 3 and 6 h. The method was found to be simple, accurate, reliable, time-saving (up to 18 samples can be determined simultaneously) and low-cost.
