Analyses of sphingosine‐1‐phosphate in the context of transfusion: how much is in stored blood products and in patient blood?

Abstract: BACKGROUND: Sphingosine-1-phosphate (S1P) is a bloodborne lipid that regulates vascular tone and endothelial permeability. S1P concentrations are reduced in critically ill patients. As hematopoietic cells produce S1P, this study intends to investigate S1P concentrations in blood products during storage and in patient plasma after blood transfusion. STUDY DESIGN AND METHODS: S1Pconcentrations were measured in 83 red blood cell (RBC) units and 73 platelet concentrates (PCs) before and after storage. In addition,… Show more

“…A concomitant increase in ceramides and decrease in sphinosine‐1‐phosphate suggests a shift in sphingolipid metabolism . Level of sphinosine‐1‐phosphate is maximum in freshly isolated platelet concentrates and decreases progressively during storage to 60% within 4 days . These observations may influence the choice of platelet concentrates for transfusion particularly in critically ill patients who show significantly reduced plasma levels of sphinosine‐1‐phosphate .…”

“…S1P levels are relatively higher in the blood than intracellularly; the concentration of serum S1P in healthy humans is approximately 1 μM [ 107 ]. S1P is stored in endothelial cells, erythrocytes, and thrombocytes [ 108 , 109 ].…”
