Analysis of 21 X-chromosomal STRs in an Algerian population sample

Abstract: Twenty-one X-chromosomal short tandem repeat loci, including the six clusters of linked markers DXS10148-DXS10135-DXS8378 (Xp22), DXS7132-DXS10074-DXS10079 (Xq12), DXS6801-DXS6809-DXS6789 (Xq21), DXS7424-DXS101 (Xq22), DXS10103-HPRTB-DXS10101 (Xq26), DXS8377-DXS10146-DXS10134-DXS7423-DXS10011 (Xq28), and the loci DXS6800 and GATA172D05 were typed in a northwestern Algerian population sample (n = 210; 104 men and 106 women). Allele and haplotype frequencies were calculated. No evidence of linkage disequilibrium… Show more

“…The five significant results from 36 pair-wise comparisons were found between DXS6854 and DXS6807, DXS6854 and DXS8377, DXS6809 and DXS8377, DXS9902 and DXS8378, and DXS7423 and DXS8377. The highest association (P<0.00001) was found between DXS9902 and DXS8378, and DXS7423 and DXS8377, which have also been reported by Bekada et al [5]. In addition, alleles of linked loci form haplotype that recombine during meioses.…”

“…The comparison of allele frequency distribution were preformed between the targeted populations and other published populations originating from Sichuan Han in China [16], Taiwanese [17], Northern Italy [4], German [18], Japanese [19], Spanish [20], Santander [6], Portugal [21], Pakistani [22], Algerian [5], Ghana [7]. There are significant differences of most loci between Han and Uigur except at the HPRTB and DXS8377, and significant differences between Han and Mongol except at the DXS9902, HPRTB, and DXS6809, and significant differences of all the nine loci between Han and other nine reported populations (seen as Electronic supplementary Table S1) except at HPRTB in Pakistanis.…”

“…In DXS10148, large alleles 40.1 and 31 were observed. Although allele 40.1 is the largest one found so far in an East Asian population [6,11,12], large alleles are fairly frequent in Algerians and Ivorians [14,21]. The largest allele so far found in East Asians was 33.1 in Korean [6,11,12].…”

“…Loci DXS10135, DXS8378, DXS7132, DXS10074, HPRTB, DXS10101, DXS10134 and DXS7423 were amplified by means of the Mentype Argus X-8 kit (Biotype GE, Dresden Germany) according to the manufacturer's instructions. The remaining X-STRs were amplified with three in-house developed PCR assays as described by Bekada et al [7]. Typing was done by capillary electrophoresis on the ABI PRISM 310 Genetic Analyzer (Applied Biosystems, Foster City, CA) in comparison to allelic ladders and control DNA from K562 and 9947A cell lines (Promega, Madison, WI) using the GeneScan and Genotyper software version 3.7 (Applied Biosystems) [8].…”
