Analysis of Bacillus megaterium lipolytic system and cloning of LipA, a novel subfamily I.4 bacterial lipase

Abstract: The lipolytic system of Bacillus megaterium 370 was investigated, showing the existence of at least two secreted lipases and a cell-bound esterase. A gene coding for an extracellular lipase was isolated and cloned in Escherichia coli. The cloned enzyme displayed high activity on short to medium chain length (C 4^C8 ) substrates, and poor activity on C 18 substrates. On the basis of amino acid sequence homology, the cloned lipase was classified into subfamily I.4 of bacterial lipases.

“…Among the producers of esterases and lipases, Bacillus as well as Ps. stutzeri have been described (Jung et al, 2003;Ruiz et al, 2002). Species of Corynebacterium have not been described as potentials producers of esterases and lipases (Hou and Jonhnston, 1992), however, in the results obtained it was observed that the isolate Coryne.…”

“…3C). However, the stepwise introduction of affinity tags decreased the amount of produced and exported recombinant protein (Malten et al 2006;Ruiz et al 2002). A one-step affinity 20 chromatography purification of His 6 -tagged TFH from the growth medium resulted in 8.3 mg protein per liter (Yang et al 2007).…”
