Analysis of Coronary Vessels in Cleared Embryonic Hearts

Abstract: Whole mount visualization of the embryonic coronary plexus from which the capillary and arterial networks will form is rendered problematic using standard microscopy techniques, due to the scattering of imaging light by the thick heart tissue, as these vessels are localized deep within the walls of the developing heart. As optical clearing of tissues using organic solvents such as BABB (1 part benzyl alcohol to 2 parts benzyl benzoate) has been shown to greatly improve the optical penetration depth that can be… Show more

“…Confocal analysis was performed as described previously [ 17 , 47 ]. Immunolabelled embryos (E9.5–11.5) were dehydrated through a methanol series before clearing with 1 part Benzyl alcohol/ 2 parts benzyl benzoate solution (BABB, Sigma) and examined by epifluorescence on an inverted LSM710 confocal system mounted on an AxioObserver Z1 microscope (Carl Zeiss Ltd, United Kingdom) [ 17 , 47 ].…”

“…Moreover, light-sheet microscopy is becoming more and more routinely-used equipment in histology 31 . The preparation of tissues for 3D histochemistry and imaging is relatively fast with a clearing procedure of 1–4 h and immunohistochemical staining for a week, whereas fluorescence signals are well-preserved, and their quenching is limited 9 , 17 . An unwanted effect of BABB clearing is tissue shrinkage.…”

“…Opacity of these tissues is mainly caused by lipid bilayers of cell membranes and not by ECM, because these tissues contain relatively small amounts of connective tissue. Clearing of ECM-rich tissues such as skin or gingiva needs an alternative approach by equalizing RIs of the different tissue compartments that are imaged 9 , 17 .…”

“…Although this research was focused on answering key biological questions or development of LSFM setups, instead of method evaluation, a few conclusions from those studies can be drawn. First, BABB was mainly used to clear petite samples such as embryonic hearts . BABB might be also used to clear and image >2.0 mm thick regions of adult mouse heart (Figure c,g), as presented by Smith et al .…”
