Analysis of cytokine gene expression in Graves' disease and multinodular goiter.

Abstract: Cytokines have a central role in the generation of an autoimmune response and can directly affect the target organ. In Graves' disease, both the infiltrating mononuclear cells and the thyroid follicular cells produce certain cytokines, but the relative contribution of each is unclear, and there are conflicting data on the exact profile of cytokines expressed within the thyroid. To clarify these issues, we used the method of reverse transcription-polymerase chain reaction amplification to analyze cytokine gene … Show more

“…Activated monocytes/macrophages secrete IL-1 and TNF, and activated lymphocytes produce IFN-y. These potent proinflammatory cytokines are expressed in the tissues of autoimmune thyroid disease [17,18], indicating that not only recruitment but also activation of mononuclear cells really takes place in vivo. This would constitute an amplification loop of the immune and inflammatory responses through a paracrine way.…”

“…Analysis of the intrathyroidal production of cytokines in such disease has revealed the expression of IFN-y, TNF-c~ and 1], IL-2, IL-6, IL-8 and IL-10 [17,18]. In addition, human thytocytes themselves have been reported to produce IL-1, IL-6, IL-8 and TGF-I~ [17][18][19], although the expression of IL-1 is controversial [18]. Subacute thyroiditis is a relatively acute *Corresponding author.…”

“…In human samples, a concomitant lymphocytic infiltrate can be observed interspersed between follicles or in the interstitial tissue. This lymphocytic infiltrate is predominantly a cytotoxic suppressor between the epithelial cells and mainly helper-inducer T cells in the interstitial tissue [1, 10]. In both cases, the morphometric evaluation identifies this issue by a relative decrease of the follicle/stroma ratio that can be confirmed by histological evaluation.…”

“…Although such thyroid-stimulating autoantibodies are pathognomonic of GD, autoantibodies to thyroid peroxidase (TPO) and thyroglobulin (Tg) are also present [1, 10]. Moreover, some GD patients have mild lymphocytic thyroiditis.…”
