Analysis of genetic diversity in oilseed brassica germplasm through ISSR markers and isozyme profiling

Abstract: Eleven Brassica germplasm were characterized through the application of 12 oligonucleotide Inter Simple Sequence Repeat (ISSR) primers. A total 1248 bands were amplified through polymorphic chain reaction and were visualized by agarose gel electrophoresis. Among the amplified marker bands 71.47% were polymorphic in nature and 352 bands were found to be monomorphic. The polymorphic bands of the amplified DNAs mostly ranged between 110 bp and 3 kb. Genetic distance among the germplasm ranged between 0.0468 and 0… Show more

“…It signifies a particular, distinct genomic DNA part that was noticed in PCR extension with a pair of definite primers. [93][94][95] These markers have great potential in plant identification at inter-and intra-specific species or population level. 93,96,97 Previously, the linkage mapping using SCAR markers has been linked with 18-carbon fatty acids in rapeseed.…”

“… 93–95 These markers have great potential in plant identification at inter- and intra-specific species or population level. 93 , 96 , 97 Previously, the linkage mapping using SCAR markers has been linked with 18-carbon fatty acids in rapeseed. Developed SCAR marker (L1L9) shows about 25% genetic difference for 18-carbon fatty acids in rapeseed.…”

“…The results were congruent with those in [ 48 ], which identified and characterized eight species of Cuscuta genes based on molecular (ITS) and morphological data and used the analysis of phylogeny to confirm the diversity among Cuscuta species. Information about genetic distance among the accessions is helpful in a breeding program [ 49 , 50 ].…”

“…In recent years, PCR-based DNA marker systems such as AFLP, RFLP, RAPD, SSR and ISSR have been common for investigating the genetic makeup of populations, genetic characterization, genetic diversity, divergence and phylogenetic studies (Li and Ge 2001;Gajera et al 2010;Chen et al 2017;Hocaoglu-Ozyigit et al 2020). ISSR markers have been proposed as a new source of genetic markers that are inherited in Mendelian fashion and are scored as dominant markers (Paul et al 2020). These markers have the role of analyzing genetic diversity through the classification of varieties (Kiani and Siahchehreh 2018).…”
