Analysis of Human Blood Plasma Proteome from Ten Healthy Volunteers from Indian Population

Abstract: Analysis of any mammalian plasma proteome is a challenge, particularly by mass spectrometry, due to the presence of albumin and other abundant proteins which can mask the detection of low abundant proteins. As detection of human plasma proteins is valuable in diagnostics, exploring various workflows with minimal fractionation prior to mass spectral analysis, is required in order to study population diversity involving analysis in a large cohort of samples. Here, we used ‘reference plasma sample’, a pool of pla… Show more

“…Bioanalysis as a subdiscipline of contemporary analytical chemistry covers quantitative as well as qualitative measurements of xenobiotics (drugs and their metabolites) and biotics (proteins, RNA, DNA) in biological matrices/systems named overall as human body fluids (HBF). For the most often mentioned HBF are blood/plasma/serum , cerebrospinal fluid , and urine . Beside those three media in the recent years also saliva as well as tears have been successfully added to analytical chemistry.…”

“…These methods are used as the first dimension of separation, either for proteins or after digestion into peptides, prior to LC-MS/MS analysis. 95,96 Furthermore, immunoaffinity-based depletion of highly abundant proteins has become the dominant approach in blood plasma proteomics to reduce the dynamic range of blood samples. Several kits and methods are available for plasma depletion via a single or multi-step procedure.…”

“…Serum/plasma provides a rich source of biological information with a dynamic range of protein abundance exceeding 10 orders of magnitude [26]. Around 24% of classical plasma proteins are associated with immune response [27]. They reflect various aspects of the functioning of adaptive and innate immune systems, as well as other related physiological processes, including wound healing, extracellular matrix remodeling, angiogenesis, complement cascades, and metabolic changes [4,28].…”
