Maqui (Aristotelia chilensis) is an edible berry native to Southern Chile. The anthocyanin profiles and concentrations of this fruit have been widely studied. The high concentration of anthocyanins confers a deep colour to the skin and pulp of maqui berries, and the biological activity of this fruit has been attributed to these compounds. However, other compounds such as flavonols and hydroxycinnamic acid derivatives in this fruit have not been studied to date.The objective was to determine the hydroxycinnamic acid derivatives and flavonol profiles in maqui berries in order expand the knowledge base for this fruit. Two extraction methodologies (solid phase extraction using a mixed mode cation exchange cartridge and selective precipitation), followed by HPLC-DAD-ESI-MS/MS were applied to the extraction and identification of the main hydroxycinnamic acids and flavonols in the berries. Quantification was accomplished by using the first extraction procedure.Low levels of hydroxycinnamic acid derivatives were detected in maqui berries, where 5-galloylquinic acid was the only quantifiable derivative. However, diverse and high levels of flavonols were detected in the fruit, the most relevant of which are myricetin-galactoside, quercetin-rutinoside, and quercetin-3-galactoside. Nineteen other flavonols were identified, with total concentrations between 1.05 and 1.18 µmol/g fresh weight.The diversity of flavonols makes the profiles and concentrations of these compounds significant, whereas hydroxycinnamic acids in maqui are not significant. The comparatively high levels of flavonols combined with the high concentration of anthocyanins contribute to the consideration of this native fruit from Chile as a "superfruit".

SUMMARYPolyphenols are chemicals found in tobacco that are affected by the method used to cure the leaf and, as a result, can be useful in the characterization of tobacco products. The purpose of this work was to develop an analytical method to investigate the levels of six polyphenols found in tobacco leaves and tobacco products: 3-O-caffeoylquinic acid (chlorogenic acid), 4-O-caffeoylquinic acid (cryptochlorogenic acid), 5-O-caffeoylquinic acid (neochlorogenic acid), kaempferol 3-O-rutinoside (nicotiflorin), quercetin 3-O-rutinoside (rutin), and 6-methoxy-7-hydroxycoumarin (scopoletin). Extraction conditions for sample preparation using PLE and instrument conditions for analysis by UPLC-MS/MS were optimized and validated. Results from the analysis of 30 cured tobacco leaves are presented and discussed in the context of each curing method represented.Results from the analysis of various tobacco products are also presented and trends observed across product types are discussed in the context of the applicability of the validated method. Total polyphenol levels for flue-cured, Oriental, and air-cured leaves were determined to be in the ranges of 18-41 mg/g, 5-27 mg/g, and 0.5-3 mg/g respectively. Similarly, cigarette polyphenol levels were found in the range of 4-16 mg/g and cigar polyphenol levels were less than 1.5 mg/g. The trends observed in the results for the tobacco leaf samples are consistent with expectations regarding the fate of polyphenols under the conditions commonly used in curing procedures. The results for the tobacco products demonstrate that the validated method can be used to study polyphenol content in cigarettes and a variety of cigar types including pipe tobacco cigars. [Beitr. Schinus lentiscifolius Marchand has been used in folk medicine to treat immunoinflammatory related diseases, which are marked by OS and altered HSR. Our study aimed to evaluate OS and HSR in lymphocytes treated with S. lentiscifolius bark extracts. S. lentiscifolius barks were partitioned with solvents to obtain hexane (SL‐HEX), ethyl acetate (SL‐ACOET) and methanol (SL‐MEOH) extracts, and the presence of bioactive compounds was evaluated by thin layer chromatography. Total phenols were measured by the Folin–Ciocalteu method and flavonoids were identified by HPLC‐DAD‐ESI‐MS/MS. Antioxidant capacity was verified by DPPH method, cell viability by Trypan Blue method, lipid peroxidation by TBARS and HSP70 by immunoblotting. The SL‐ACOET extract presented higher content of phenolic compounds and antioxidant activity in vitro. It was able to reduce lipid peroxidation levels in lymphocytes induced by H2O2 and improved cell viability. The SL‐ACOET extract inhibited HSR by a decrease in both intracellular content and release of 70 kDa heat shock proteins (HSP70) and also by decrease extra‐to‐intracellular HSP70 ratio in lymphocytes submitted to heat shock (2 h, 41 °C). S. lentiscifolius bark extract has antioxidant activity and inhibitory effect on HSR probably due to the presence of polyphenols as the flavonoids quercetin and kaempferol.
