Analysis of initiation of sites of cell wall growth in Streptococcus faecium during a nutritional shift

Abstract: Cell counts. At intervals, 0.1-ml samples of cultures were added to 0.4 ml of 10.4% Formalin at room temperature. After thorough agitation, the cells were kept at room temperature for at least 30 min but not more than 6 h. Then samples of 0.05 or 0.10 ml of the Formalin-fixed cell suspension were added to a 30-ml plastic cup, and a volume of 0.9% sterile saline was added to dilute the samples, so that between 5,000 and 15,000 cells were analyzed per sample.
