Analysis of natural product regulation of cannabinoid receptors in the treatment of human disease

Abstract: The organized, tightly regulated signaling relays engaged by the cannabinoid receptors (CBs) and their ligands, G proteins and other effectors, together constitute the endocannabinoid system (ECS). This system governs many biological functions including cell proliferation, regulation of ion transport and neuronal messaging. This review will firstly examine the physiology of the ECS, briefly discussing some anomalies in the relay of the ECS signaling as these are consequently linked to maladies of global concer… Show more

“…Hence, the synergistic anti‐tumor effect of Alantolactone could help to overcome the drug resistance and sensitize the EGFR targeted therapy for PC. Although, Alantolactone has good safety and pharmacological properties as reported before, the effect of pharmacokinetics are poor. The next step we will study the nanotechnology‐based therapeutics, which could improve the Alantolactone bioavailability and tumor targetability.…”

“…Recent studies have drawn attention to the antitumor properties of natural products due to their confirmed pharmacological properties and few side effects . Alantolactone acts as an agent with active antitumor activity, which inhibits tumor cell proliferation, promotes apoptosis, and suppresses invasion and metastasis .…”

“…A number of studies have investigated CBD to determine its activity at cannabinoid receptors and shown very low affinity for CB1 and CB2 [ 14 ] ( Table 1 b). It was reported that CBD can act as an antagonist/inverse agonist at certain concentrations below which it binds to both CB1 and CB2 orthosteric sites [ 53 ]. Recently, several studies demonstrated that CBD can act as a negative allosteric modulator of CB1, which alters the potency and efficacy of the orthosteric ligands but does not activate the receptor itself [ 54 , 55 , 56 ].…”

“…Reseptor ini terletak di beberapa bagian tubuh terutama di otak (Mackie, 2008). Reseptor canabinoid bersama dengan ligan membentuk sistem endocannabinoid (endocannabinoid system) yang dimodulasi oleh senyawa yang terdapat pada tanaman darat dan air termasuk terpena (terpenoid) yang terkandung pada tanaman obat kipahit dan jambu biji (Badal et al, 2017).…”
