Analysis of peripheral blood for prostate cells after autologous transfusion given during radical prostatectomy

Abstract: peripheral blood samples were collected from patients before RP, in the recovery room afterward, and at 3-5 weeks after surgery. A reverse-transcriptase-polymerase chain reaction assay for PSA mRNA was used to detect prostate cells in cell-saver and peripheral blood samples. Patients were followed after surgery with PSA measurements to assess biochemical failure. RESULTSPSA-expressing cells were detected in 88% of cell-saver reservoir and 13% of preoperative blood samples. No PSA-expressing prostate cells were… Show more

“…[19] They used a reverse transcriptase polymerase chain reaction (RT-PCR) for their evaluation and found these cells in 88% of the patients in the immediate postoperative period. When these patients were tested at 6-week interval, they did not find any PSA-expressing cells and neither did they demonstrate any increased biochemical recurrence following the use of ICS.…”

“…23 Stoffel et al used a reverse transcription polymerase chain reaction assay for PSA mRNA to detect prostate cells in the cell-savaged and peripheral blood samples of patients during and following radical prostatectomy. 24 Although PSA expressing cells were found in the ICS blood of the majority of patients, they could not detect it in the peripheral circulation of any patients 3-5 weeks postoperatively with no biochemical failures.…”

“…In these studies, blood was collected from the surgical field subsequent to prostatic manipulation. Stoffel performed measurements of the Prostate Specific Antigen (PSA) in subjects' blood: 1 hour prior to surgery; immediately subsequent to reinfusion of the shed blood; and 3-5 weeks following surgery (Stoffel et al 2005). He also measured the PSA level in the processed salvaged blood.…”

“…Autotransfusion of washed unradiated shed blood is often employed during cystectomies and prostatectomies (Nieder et al 2004, Davis et al 2003, Ford et al 2007, Gallina et al 2007, Stoffel et al 2005, Waters et al 2004). In these studies, blood was collected from the surgical field subsequent to prostatic manipulation.…”
