Barbarea integrifolia DC. (Brassicaceae) is an endangered and endemic species located in Erzincan and Gümüşhane provinces of Turkey. In total, 27 individuals from 2 natural populations were assessed using the random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) method coupled with sequence analysis of the internal transcribed spacer (ITS1) rDNA region. Genetic accuracy of RAPD-PCR was tested with 25 RAPD primers, and it resulted in 115 clear and reproducible DNA fragments from 13 RAPD primers. Among these, 76 (66.1%) fragments were found to be polymorphic. According to the ITS1 analysis of B. integrifolia populations, the plants exhibited a relatively poor genetic diversity. In total, 60 ITS1 nucleotide sequences from the GenBank database with 2 newly identified ITS1 sequences from the Turkish populations were used to construct a maximum-likelihood tree. The lengths of ITS1 sequences of B. integrifolia were found to be 268 bp for population I and 271 bp for population II. ITS1 sequences of B. integrifolia species showed 97% sequence identity. Phylogenetic analysis revealed that Barbarea species were monophyletic and showed high bootstrap values. B. integrifolia had closer relationships with Cardamineae taxa, including Cardamine, Rorippa, and Armoracia. On the contrary, allied genus Nasturtium species were separated from Cardamineae taxa. The genetic information obtained from this study could be used for the development of conservation strategies not only for B. integrifolia but also for rare and endangered plant species.

Summary• A hallmark of the innate immune system of plants is the biosynthesis of lowmolecular-weight compounds referred to as secondary metabolites. Tryptophanderived branch pathways contribute to the capacity for chemical defense against microbes in Arabidopsis thaliana.• Here, we investigated phylogenetic patterns of this metabolic pathway in relatives of A. thaliana following inoculation with filamentous fungal pathogens that employ contrasting infection strategies.• The study revealed unexpected phylogenetic conservation of the pathogeninduced indole glucosinolate (IG) metabolic pathway, including a metabolic shift of IG biosynthesis to 4-methoxyindol-3-ylmethylglucosinolate and IG metabolization. By contrast, indole-3-carboxylic acid and camalexin biosyntheses are clade-specific innovations within this metabolic framework. A Capsella rubella accession was found to be devoid of any IG metabolites and to lack orthologs of two A. thaliana genes needed for 4-methoxyindol-3-ylmethylglucosinolate biosynthesis or hydrolysis. However, C. rubella was found to retain the capacity to deposit callose after treatment with the bacterial flagellin-derived epitope flg22 and pre-invasive resistance against a nonadapted powdery mildew fungus.• We conclude that pathogen-inducible IG metabolism in the Brassicaceae is evolutionarily ancient, while other tryptophan-derived branch pathways represent relatively recent manifestations of a plant-pathogen arms race. Moreover, at least one Brassicaceae lineage appears to have evolved IG-independent defense signaling and ⁄ or output pathway(s).

Molecular phylogeny and systematics of the tribe Chorisporeae (Brassicaceae)

Sequence data from nuclear (ITS) and chloroplast (trnL-F) regions for 89 accessions representing 56 out of 64 species from all five genera of the tribe Chorisporeae (plus Dontostemon tibeticus) have been studied to test the monophyly of the tribe and its component genera, clarify its boundaries, and elucidate its phylogenetic position in the family. Both data sets showed strong support for the monophyly of the Chorisporeae as currently delimited, though the position of its tentative member D. tibeticus was not resolved by ITS. Parrya and Pseudoclausia are polyand paraphyletic with regard to each other, and Chorispora is either polyphyletic or at least paraphyletic (comprising Diptychocarpus) within a weakly supported monophyletic clade. The incongruence in branching pattern among the markers was most likely caused by hybridization and possibly influenced by incomplete lineage sorting. The present results suggest uniting Pseudoclausia, Clausia podlechii, and Achoriphragma with Parrya and transferring P. beketovii and P. saposhnikovii to Leiospora (Euclidieae). We also obtained support for splitting Chorispora into two geographically defined groups, one of which is closer to Diptychocarpus. Both data sets revealed a close relationship of the Chorisporeae to Dontostemoneae, while ITS also indicated affinity to Hesperideae. Therefore, the position of Chorisporeae needs further verification.
