Analysis of Reparative Activity of Platelet Lysate: Effect on Cell Monolayer Recovery In Vitro and Skin Wound Healing In Vivo

Abstract: Platelet lysate prepared from donor platelet concentrate and pooled according to a developed technique stimulates migration of multipotent mesenchymal stromal cells of the human adipose tissue and promotes healing of the monolayer defect in cultures of human fibroblasts and multipotent mesenchymal stromal cells in vitro in concentrations close those of fetal calf serum (5-10%). Lysate of platelets from platelet-rich rat blood plasma stimulated healing of the skin defect by promoting epithelialization and granu… Show more

“…24 Platelet lysate has been shown to have effects on cell monolayer recovery in vitro and in the setting of wound healing in vivo. 25 We speculated that functional factors that promote wound healing may be decreased during pregnancy due to internal structural changes of platelets because CSD prevalence is negatively correlated with post-CS platelet concentration. 26 Although a higher fibrinogen concentration is beneficial in vaginal delivery, it is a predictor for CSD in women undergoing CS.…”

“…If a microscopic tumor is suspended in dormancy by insufficient blood supply, this release of VEGF may constitute an "angiogenic switch" allowing escape from dormancy by stimulated angiogenesis [17,30] and VEGF levels have been correlated with disease progression [31]. In vitro and in vivo studies have demonstrated both improved wound healing and enhanced breast cancer cell colony formation by addition of platelet lysate [32,33,34]. The main effect of platelet activation on cancer seems to be to promote invasion and metastasis.…”
