Analysis of simple sequence repeat markers in homozygous lines of apple

Abstract: Haploids in apple were induced using anther culture and in situ parthenogenesis followed by in vitro culture of immature embryos or cotyledons. In addition to an earlier characterization of the zygosity state using isozymes, the purpose of this work was an analysis of simple sequence repeats (SSRs) to describe the homozygosity more comprehensively. Five previously described SSR primer combinations were applied. Initially, polymerase chain reaction amplification with the primer pairs was performed on the donor … Show more

“…This somaclonal variation has been detected with RAPDs (Raimondi et al 2001), producing a changes similar to the obtained by Eimert et al (2003). The EST-SSRs have been used to determine the origin of the regenerated plantlets from anther culture in different species such as rice (Lapitan et al 2009), apple (Höfer et al 2002;Vanwynsberghe et al 2005;Okada et al 2009), corn (Aulinger et al 2003), soy (Rodrigues et al 2004), potato (Veilleux et al 1995;Chani et al 2000), coconut (Perera et al 2008), orange (Cao et al 2011) or eggplant (Salas et al 2011).…”

“…Mendelian inheritance and high polymorphism make nuclear SSR markers very efficient for the evaluation of heterozygosity (vs homozygosity) in diploids (Chani et al, 2000;Hofer et al, 2002;Fei Yu et al, 2004;Vanwynsberghe et al, 2005). However, null alleles are frequently observed and have to be considered for genetic analysis (Dakin and Avise, 2004;Hoffman and Amos, 2005).…”

“…Triploids regenerated from anther culture have been reported in apple (Ho¨fer, 1994;Ho¨fer et al, 2002). Isozyme analyses have been employed to confirm the gametic origin of calluses and plantlets in apple (Ho¨fer and Grafe, 2000).…”
