Analysis of Surface-Exposed Outer Membrane Proteins in Helicobacter pylori

Abstract: More than 50Helicobacter pylori genes are predicted to encode outer membrane proteins (OMPs), but there has been relatively little experimental investigation of the H. pylori cell surface proteome. In this study, we used selective biotinylation to label proteins localized to the surface of H. pylori, along with differential detergent extraction procedures to isolate proteins localized to the outer membrane. Proteins that met multiple criteria for surface-exposed outer membrane localization included known adhes… Show more

“…Secretion of VacA is believed to proceed via a type V or auto-transporter mechanism (6,(34)(35)(36)(37)(38). The secreted 88-kDa VacA protein can undergo limited proteolysis to yield two domains, designated p33 and p55, which remain together after proteolysis (27,36,(39)(40)(41)(42).…”

“…The third ring from the outside shows the GC-skew between the genomes. The cyan lines mark the distribution of outer membrane protein (OMP) genes (as defined by Alm et al, 2000 and Voss et al, 2014). (B) A Venn diagram compares the core and pan-genomes of Nic_25A and P12 based on orthologous clustering with 0.8 identity cut-off.…”

“…As seen in the Figure 4 , the adhesion factors AlpB, HpaA, and HopQ were also found to be more abundant in the P12 strain. Based on the unique proteins identified in the triplicate runs ( Supporting File 8 ), the adhesins BabB and SabB were more abundant in the P12 strain, as were OMPs of the Hop and Hom families (Voss et al, 2014) and other virulence factors including vacuolating cytotoxic protein A (VacA), elongation factor (EF-Tu), and flagellar basal body protein FliL.…”

“…Bacterial cultures were harvested by centrifugation at 3,500 ϫ g, and the pellets were resuspended in phosphate-buffered saline. Following a second centrifugation at 3,500 ϫ g, pellets were resuspended in Tris buffer containing cOmplete protease inhibitor cocktail (Roche) as described previously (29). Bacteria were lysed by sonication, and lysates were clarified by centrifugation.…”

“…Bacteria were lysed by sonication, and lysates were clarified by centrifugation. These preparations were then analyzed by multidimensional protein identification technology (MudPIT) as described previously (29)(30)(31). Peptide tandem mass spectra were queried with SEQUEST (full tryptic specificity) against an H. pylori strain B8 (a closely related strain for which a complete genome sequence is available) (32) database to which both common contaminants and reversed versions of the proteins had been appended.…”
