Analysis of type-restricted and cross-reactive epitopes on virus-like particles of human papillomavirus type 33 and in infected tissues using monoclonal antibodies to the major capsid protein

Abstract: A panel of six monoclonal antibodies recognizing at least three different antigenic regions has been raised against the L1 major capsid protein of human papillomavirus type 33 (HPV-33), which is associated with cervical carcinoma. The antigenic sites defined by these antibodies have been mapped and classified as typerestricted or broadly cross-reactive using bacterially expressed L1 fusion proteins of a variety of HPV types.Conformational and linear epitopes have been distinguished using native and denatured v… Show more

“…For detection of V5-tagged OBSL1 after Western blotting or immunofluorescence staining, either monoclonal antibody (MAb) SV5-Pk1 (Bio-Rad, Hercules, CA) or polyclonal antibody ab9116 (Abcam, Cambridge, UK) was used. HPV16 L1-specific mouse monoclonal antibody 16L1-312F and rabbit polyclonal antibody K75 and 33L1-7 as well as HPV16 L2-specific mouse antibody 33L2-1 have been previously described (L2-1 [60], K75 [61], 312F [62], and L1-7 [63]). Obscurin-like 1-specific polyclonal rabbit antibody HPA036404 and FLAG (M2)-and ␤-actin (A5441)-specific mouse antibodies were purchased from SigmaAldrich, St. Louis, MO.…”

“…The Bromo-2=-deoxy-uridine (BrdU) Labeling and Detection Kit I was from Roche. The L1-7 and RG-1 monoclonal antibodies were kind gifts from M. Sapp (Louisiana State University, Shreveport, LA, USA) (36) and from R. Roden (Johns Hopkins University, Baltimore, MD, USA) (37), respectively. The CAMVIR-1 and LAMP1 (H4A3) antibodies were from Santa Cruz Biotechnology, the monoclonal kallikrein-8 (KLK8) rabbit antibody (EPR5752) (2) was from Abcam, the TGN46 antibody (AHP1586) was from Abd Serotec, and the tubulin antibody (DM1A) was from Sigma.…”

“…All cells were grown in Dulbecco modified Eagle medium (DMEM) supplemented with 10% fetal calf serum and antibiotics at 37°C. L1-and L2-specific mouse monoclonal antibodies (MAbs) and rabbit polyclonal antibodies were as described elsewhere (29,37,38). Daxxspecific antibody was purchased from Santa Cruz.…”
