Analytical Methods for Determination of Counter-ions in Pharmaceutical Salts

Abstract: Inorganic ions as well as organic acids or bases are frequently employed as counter-ions of ionizable drug substances. Therefore, analytical methods are required to confirm production of the correct salt form and to quantify the level of the counter-ion present in each batch of drug substances manufactured. Several options exist for determination of counter-ions in pharmaceutical salts and will be discussed in this review. Ion chromatography (IC) with conductivity detection is probably the most frequently empl… Show more

“…Separating and monitoring inorganic ions are extremely important, with applications in many fields including: environmental [1,2], pharmaceutical [3,4], forensics [5,6], and defense, such as identifying the chemicals used in improvised explosive devices [7,8]. Conventional analytical methods for inorganic ion and speciation analysis include HPLC, ion chromatography (IC), atomic absorption spectrometry, atomic emission spectroscopy, and ICP-MS [9].…”

“…1) Development of a salt formulation is one of the most popular techniques to improve physicochemical properties of a compound, and many drugs having a salt formulation as drug substance are approved. [2][3][4][5][6][7] However, salt forms are not adopted to neutral compounds, since they require that the compound has some dissociative group. 8) In addition, the number of counter molecules that can be used to form salts is small, because their safety information and approval records as salts are considered in use as pharmaceutical salts.…”
