Anandamide Capacitates Bull Spermatozoa through CB1 and TRPV1 Activation

Abstract: Anandamide (AEA), a major endocannabinoid, binds to cannabinoid and vanilloid receptors (CB1, CB2 and TRPV1) and affects many reproductive functions. Nanomolar levels of anandamide are found in reproductive fluids including mid-cycle oviductal fluid. Previously, we found that R(+)-methanandamide, an anandamide analogue, induces sperm releasing from bovine oviductal epithelium and the CB1 antagonist, SR141716A, reversed this effect. Since sperm detachment may be due to surface remodeling brought about by capaci… Show more

“…In human sperm cells, both hot and cold-sensitive TRP ion channels are present and regulate several functions related to motility and fertility. [26][27][28] In boar sperm cells, TRPV1 function is important for fertilization. [29][30][31] In mouse sperm cells, TRPP2 is present at the anterior sperm head region and it is essential for the zona pellucida induced Ca 2+ entry into mouse sperm and subsequent acrosomal exocytosis during fertilization.…”

“…Spermatozoa also express the vanilloid TRPV1 receptor. Along with CB1 receptors, the TRPV1 receptor has been found to play a role in spermatozoa capacitation [71]. The activation of the TRPV1 receptor through AEA binding helps to prevent the spontaneous acrosome reaction to occur in an untimely manner before reaching the oocyte.…”

“…The activation of the TRPV1 receptor through AEA binding helps to prevent the spontaneous acrosome reaction to occur in an untimely manner before reaching the oocyte. Unlike the CB1 and CB2 receptors, binding to the TRPV1 receptor occurs intracellularly [71,72].…”

“…FAAH is bound to intracellular membranes [45] . Obstruction AEA degradation by URB97 (an inhibitor of FAAH) caused sperm capacitation and the pre-incubation of sperm with CB1 or TRPV1 antagonists inverted this result [46] . It is proposed that cellular uptake of AEA can be controlled with FAAH by maintaining and/or creating an inward concentration gradient that drives the facilitated diffusion of AEA through anandamide membrane Transporter (AMT).…”
