Androgens and ageing of the skin

Abstract: Consequently, skin becomes susceptible to infections and several disease states. Androgen treatment has been proposed to be an effective way to reverse the age-associated deterioration of skin; however, further research is needed to translate current findings to clinically significant diagnostic and therapeutic applications.

“…One of the key enzymes, cytochrome P450c17, which is necessary for synthesis of dehydroepiandrosterone and androstenedione, has been documented to be expressed in human sebocytes. These androgenic pro-hormones can be further converted by sebocytes and sweat glands, and probably also by dermal papilla hair cells, into more potent androgens, such as testosterone and 5α-DHT (Makrantonaki and Zouboulis, 2009;Zouboulis et al, 2007). In addition, the enzyme aromatase may convert testosterone and androstenedione to estrogens in sebaceous glands, outer as well as inner root sheath cells of anagen terminal hair follicles, and dermal papilla cells (Fritsch et al, 2001;Thornton et al, 2006).…”

“…Estrogen deficiency directly affects skin homeostasis [3] and may also result in dysregulation of peripheral androgen metabolism. Androgens have been documented to influence skin morphology and play a distinguished role in epidermal barrier homeostasis, wound healing, sebaceous gland growth, differentiation and hair growth [4]. …”
