Scope Sirtuin 3 (SIRT3) plays a protective role against nonalcoholic fatty liver disease (NAFLD) by improving hepatic mitochondrial dysfunction. Gut microbiota imbalance contributes to the pathogenesis of NAFLD, yet the underlying mechanism linking SIRT3 with gut microbiota in NAFLD progression remains obscure. Methods and results Wild‐type 129 mice and SIRT3 knockout (SIRT3KO) mice are placed under a chow diet or high‐fat diet (HFD) treatment for 18 weeks. HFD resulted in a significantly increased hepatic steatosis and inflammation, which are exacerbated in SIRT3KO mice. The gut microbiota by 16s rRNA gene sequencing and phylogenetic reconstruction of unobserved states analysis are characterized. Lack of SIRT3 facilitates gut microbial dysbiosis in mice following HFD, with increased Desulfovibrio, Oscillibacter, and decreased Alloprevotella. SIRT3 deficiency resulted in an impaired intestinal permeability and inflammation in HFD‐fed mice, which can be attenuated by sodium butyrate (NaB). SIRT3KO HFD‐fed mice is followed by an increased lipopolysaccharide into the circulation and dysregulated expressions of cannabinoid receptor 1 and 2 in colon and liver, which are significantly associated with the alterations of intestinal microbiota. Conclusions SIRT3 deficiency promotes NAFLD progression in correlation with impaired intestinal permeability through gut microbiota dysbiosis.

Comprehensive Analysis of the Global Protein Changes That Occur During Salivary Gland Degeneration in Female Ixodid Ticks Haemaphysalis longicornis

Ticks are notorious blood-sucking arthropods that can spread a variety of pathogens and cause great harm to the health of humans, wildlife and domestic animals. The salivary glands of female ticks degenerate rapidly when the ticks reach critical weight or become engorged, which can be caused by hormones and by the synergistic effects of multiple proteins. To explore the complex molecular mechanisms of salivary gland degeneration in ticks, this study applies iTRAQ quantitative proteomic technology for the first time to study changes in protein expression in the salivary glands of female Haemaphysalis longicornis during the process of degeneration and to search for proteins that play an important role in salivary gland degeneration. It was found that the expression of some proteins associated with energy production was continuously down-regulated during salivary gland degeneration, while some proteins associated with DNA or protein degradation were consistently up-regulated. Furthermore, the expression of some proteins related to cell apoptosis or autophagy was also changed. These proteins were knocked down by RNAi to observe the phenotypic and physiological changes in female ticks. The results showed that the time required for engorgement and the mortality rates of the female ticks increased after RNAi of F0F1-type ATP synthase, NADH-ubiquinone oxidoreductase, cytochrome C, or apoptosis-inducing factor (AIF). The corresponding engorged weights, oviposition amounts, and egg hatching rates of the female ticks decreased after RNAi. Interference of the expression of AIF in engorged ticks by RNAi showed that the degeneration of salivary glands of female ticks was slowed down.

Background Human neutrophil elastase (HNE) is an abundantly expressed neutrophil serine protease that promotes neutrophil invasion and neutrophil extracellular trap (NET) formation, thereby mediating lung tissue destruction and enhancing pulmonary inflammation in acute lung injury (ALI). Chemical agents that target HNE and manipulate HNE level homeostasis are desired to prevent or treat ALI. Methods In a search for HNE inhibitors, traditional Chinese medicines were evaluated, leading to the identification of the water extract of spikes of Prunella vulgaris as an inhibitor of HNE activity. Using bioactivity-guided fractionation, a bioactive fraction (PVAP) that inhibited NHE activity was prepared, and its effects on lipopolysaccharide (LPS)-induced ALI were studied. Results The results suggested that PVAP elicited protective effects against LPS-induced ALI by inhibiting HNE activity, thereby reducing neutrophil invasion, pro-inflammatory cytokine release, and NET formation. Conclusions These findings illustrated the utility of PVAP as an agent for preventing ALI.
