Aneuploidy of stem cells isolated from human adipose tissue

Abstract: The incidence of autosomes 6 and 8 aneuploidy in stem cell cultures derived from adipose tissue was evaluated at different stages of culturing. Monosomy was more incident than trisomy during the early passages. Distribution of cultures by the incidence of aneuploidy in different chromosomes was virtually the same. Clones with chromosome 6 monosomy were detected in two cultures during late passages.

“…These fi ndings, our previous results [1,2], and pub lished data [8,10] disagree with the opinion of some scientists on genetic stability of SC and safety of their use without cytogenetic testing. The data on different types of chromosome changeability in cultured MSC necessitate more profound studies of SC genetics and creation of a system of their quality control for prevention of untoward consequences of therapeutic use of these cells.…”

“…Monosomic nuclei predominated over trisomic ones. These data are in line with our previous data on MSC isolated from the adipose tissue [1]. The incidence of the studied autosome aneuploidy did not change over the course of MSC culturing (p>0.05; Table 3).…”

“…According to our data, abnormal clones were detected in about 30% adipose tissue MSC and in 11% bone marrow MSC cultures [1,2]. Initial aspirated bone mar row or adipose tissue can be the sources of ab normal clones.…”

“…The problem of evaluating clone formation in mo saic cultures, discussed previously [2], deserves special attention. The presence of a clone in a culture is confi rmed by the presence of a group of cells with the same chromosome abnormality, but the problemwhich, specifi cally, incidence of aneuploid cells in the culture implies that this culture should be classifi ed as mosaic culture with clones -deserves a special study.…”
