Angucycline Glycosides from an Intertidal Sediments Strain Streptomyces sp. and Their Cytotoxic Activity against Hepatoma Carcinoma Cells

Abstract: Four angucycline glycosides including three new compounds landomycin N (1), galtamycin C (2) and vineomycin D (3), and a known homologue saquayamycin B (4), along with two alkaloids 1-acetyl-β-carboline (5) and indole-3-acetic acid (6), were identified from the fermentation broth of an intertidal sediments derived Streptomyces sp. Their structures were mainly established by IR, HR-ESI-MS, 1D and 2D NMR techniques. Among the isolated angucyclines, saquayamycin B displayed potent cytotoxic activity against hepat… Show more

“…A slightly higher cytotoxicity was observed in HepG2 and plc-prf-5 hepatocellular carcinoma cells, where saquayamycin B displayed an IC 50 of 0.14 and 0.24 μM ( Table 2 ). On human T-lymphoblastic Jurkat cells, but also on SMMC-7721 hepatoma cells, instead, the IC 50 of saquayamycin B was almost one order of magnitude lower than those obtained on the hepatic and breast cancer cell lines ( Table 2 ) [ 120 , 123 ]. Notably, on SMMC-7721 cells, saquayamycin B (0.025–0.100 μM) dose-dependently promoted apoptosis [ 120 ].…”

“…Based on that, 7 was identified as galtamycin C, which has previously been isolated from an intertidal sediments-derived Streptomyces sp. 32 . As for 8, it was found to be 1-oxo-3-hydroxy-3,4-dihydro-2H-galtamycin C based on the different 2D NMR correlations observed in comparison with 7, together with the molecular formula and degrees of unsaturation obtained from its accurate mass (Supplemental data).…”
