Anti-allergic effects of natural tetranortriterpenoids isolated from Carapa guianensis Aublet on allergen-induced vascular permeability and hyperalgesia

Abstract: Objective: We investigated the anti-allergic and analgesic properties of an oil and a derived fraction of tetranortriterpenoids (TNTP) obtained from the seeds of Carapa guianensis Aublet. Materials and methods: Pleurisy, paw and ear edema were induced in Swiss and C57/Bl10 mice mice, whereas thermal hyperalgesia was assessed in Wistar rats (n = 6 -10 per group). Values of p < 0.05 were regarded as significant. Results: C. guianensis oil (100 to 400 mg/kg, p. o.) and TNTP (12.5 to 100 mg/kg, p. o.) inhibited pl… Show more

“…Our group previously demonstrated that gedunin presents important anti-inflammatory and immunomodulatory effects in different experimental models in vivo, including experimental arthritis, allergic pleurisy, and allergic lung inflammation, in which macrophages play a pivotal role (Penido et al, 2005(Penido et al, , 2006aHenriques and Penido, 2014;Conte et al, 2015). Here, we demonstrate that gedunin directly modulates macrophages in vitro, in a concentration-dependent manner, inhibiting early and classic parameters of macrophage activation after pathogen-associated molecular pattern recognition.…”

“…The plant anti-inflammatory properties are thought to depend on the presence of limonoids, as testified by studies on murine models of arthritis and allergy [27,28]. Edema formation has been inhibited by the oil in different rodent models via the impairment of signaling pathways triggered by histamine, bradykinin, and platelet-activating factor [29].…”

“…Other studies proved that this oil has analgesic, anti-allergic, and antihistaminic activity, inhibiting the formation of local edema (PENNAFORTE, 2003). Antiedema and analgesic properties of andiroba oil and a derived fraction of this oil, rich in tetranortriterpenoids, were confirmed in Swiss C57/BL10 strain mice with induced pleurisy and edema of the limbs (PENIDO et al, 2005) and zymozan-induced arthritis (PENIDO et al, 2006).…”

“…In the present study, even with administration of NSAID, there was local pain reaction more evident in GO and GC wounds on the 3 rd day of treatment. This may be explained by andiroba oil's analgesic effects due to blockage of inflammatory mediators (eicosanoids) (PENIDO et al, 2005). Although proposed protocol for the evaluation of local pain through behavioral analysis is considered subjective, blinded examination prevented that recorded results be influenced.…”
