Anti-Amyloid Aggregation Activity of Black Sesame Pigment: Toward a Novel Alzheimer’s Disease Preventive Agent

Abstract: Black sesame pigment (BSP) represents a low cost, easily accessible material of plant origin exhibiting marked antioxidant and heavy metal-binding properties with potential as a food supplement. We report herein the inhibitory properties of the potentially bioaccessible fraction of BSP following simulated gastrointestinal digestion against key enzymes involved in Alzheimer’s disease (AD). HPLC analysis indicated that BSP is transformed under the pH conditions mimicking the intestinal environment and the most a… Show more

“…P-coumaric acid (OB = 43.29, DL = 0.04) can relieve the neuroinflammatory responses induced by amyloid-beta peptide [ 61 ]. Vanillic acid exhibits marked antioxidant properties [ 62 ]. P-MCA (p-methoxycinnamic acid, OB = 31, DL = 0.05) exerts antihyperglycemic [ 63 ] and neuroprotective effects [ 64 ].…”

“…Moreover, a novel composition of palm oil, which is rich in PCA and 4-hydroxybenzoic acid, with lesser amounts of syringic, gallic, and vanillic acids, has been proposed for the treatment of Alzheimer’s disease based on its ability to inhibit amyloid beta fibril formation [205]. Extracts rich in vanillic acid from cyanobacteria and biotransformed black sesame seed pigment have also shown inhibitory activity towards amyloid beta aggregate formation, and were reported to significantly reduce the activity of beta-secretase and prevent the toxicity of such aggregates [184,206]. In a similar fashion, both PCA and GA were found to inhibit the formation of toxic α-synuclein fibrils, destabilize pre-formed fibrils of this protein, and preserve cellular viability in a cell line treated with α-synuclein oligomers and protofibrils [192,204,207].…”
