Anti‐inflammatory action of ark shell ( Scapharca subcrenata ) protein hydrolysate in LPS‐stimulated RAW264.7 murine macrophages

Abstract: Potential anti‐inflammatory effects of ark shell (Scapharca subcrenata) protein hydrolysates were investigated. Ark shell protein hydrolysates were prepared using Alcalase® and pepsin and were designated ASAH and ASPH, respectively. The nitric oxide (NO) inhibitory activity of ASAH and ASPH was determined in lipopolysaccharides (LPS)‐stimulated RAW264.7 murine macrophages, and the results showed that ASAH inhibited better NO inhibitory activity than ASPH. ASAH suppressed inflammatory mediator, a prostaglandin … Show more

“…Apart from their edible value, both ark clams have been found to have beneficial physiological effects. Peptides from the protein hydrolysate of SS and TG were reported to have anti‐inflammatory 17,18 and antioxidant 19,20 effects. Additionally, our previous studies have shown that dietary supplementation with the two ark clams could ameliorate endothelial dysfunction in rats 21 .…”

“…The levels of p-NF-κB protein in LPS-induced cells fell by 13.5%, 48.0%, and 52.0% when treated with 5, 10, and 20 g/mL of FLs, respectively. Blocking the activation of extracellular signal-regulated kinase (ERK) and P38 in the MAPK signaling pathway is another effective means to suppress inflammation [37,38]. Compared to the control group, the protein levels of p-P38 and p-ERK in the LPS group were enhanced 22.9-fold and 18.3-fold, respectively (Figure 3A).…”

“…Weight Distribution and Amino Acid Composition. Te molecular weight distribution and amino acid composition of ASPH were determined as described by Marasinghe et al [30]. Molecular weight distribution was analyzed using the HPLC system (Dionex UltiMateTM 3000, Termo Scientifc) equipped with a Superdex Peptide 10/300 GL column (No.…”

“…Association of Ofcial Analytical Chemist (AOAC) standard protocols were followed to analyze the proximate composition of ASPH according to the method of Marasinghe et al [30]. Briefy, the method 988.12 (phenol-sulfuric acid method), method 922.06, method 923.03, and method 925.10 were used to determine crude carbohydrate content, crude lipid, crude ash, and moisture content, respectively.…”
