Anti-inflammatory Actions of Herbal Formula Gyejibokryeong-Hwan Regulated by Inhibiting Chemokine Production and STAT1 Activation in HaCaT Cells

Abstract: Gyejibokryeong-hwan (GJBRH; Keishi-bukuryo-gan in Japan and Guizhi Fuling Wan in China) is a traditional herbal formula comprising five medicinal herbs and is used to treat climacteric syndrome. GJBRH has been shown to exhibit biological activity against diabetes, diabetic nephropathy, atherosclerosis, ischemia, and cancer. However, there is no scientific evidence of its activities against skin inflammation, including atopic dermatitis. We used the HaCaT human keratinocyte cell line to investigate the effects … Show more

“…Amygdalin (AG), a natural product derived from the aromatic amino acid phenylalanine, has a lot of cell-protective functions in many cell lines [18][19][20]. Previous studies have suggested that AG has multiple pharmacological effects, including anticancer, antibiotic, and anti-inflammation activities [21,22]. Recent studies [23] have reported that AG markedly improved glucose tolerance, and ameliorates insulin action in obese and/or diabetic patients by activating a signalling pathway related to glucose metabolism.…”

“…Our data are consistent with those of previous studies showing that the phosphorylation of STAT1 is responsible for the anti-inflammatory effect in cells treated with several herbs. 37,38 Finally, we identified the active compound responsible for the anti-inflammatory effect of the aerial parts of S. chinensis. Previously, sauchinone has been isolated from the n-hexane layer of S. chinensis roots and reported to induce HO-1 expression via the ERK signaling pathway in RAW264.7 cells.…”

“…Thus, it is necessary to develop anti-inflammatory drugs with improved efficacy against skin inflammation, with fewer side effects, that may be used over longer periods of time. Recently, various studies have reported on the inhibitory effects of traditional herbal formulas on skin inflammation (14)(15)(16)(17)(18) and have indicated that herbal formulas may be valuable alternative medicines for treating skin inflammation diseases. Hyangso-san (HSS) is a traditional herbal formula that has been used in Korea to treat symptoms of the common cold, including headache, pantalgia, fever and chills (19).…”
