Cultural and economic shifts in the early 19th century led to the rapid development of companies that made good profits from technologically-produced commodities. In this way, some habits changed in society, such as the overconsumption of processed and micronutrient-poor foods and devices that gave rise to a sedentary lifestyle. These factors influenced host-microbiome interactions which, in turn, mediated the etiopathogenesis of “new-era” disorders and diseases, which are closely related, such as obesity, type 2 diabetes mellitus, non-alcoholic fatty liver disease, hypertension, and inflammatory bowel disease, which are characterized by chronic dysregulation of metabolic and immune processes. These pathological conditions require novel and effective therapeutic approaches. Morinda citrifolia (noni) is well known as a traditional healing plant due to its medicinal properties. Thus, many studies have been conducted to understand its bioactive compounds and their mechanisms of action. However, in obesity and obesity-related metabolic (dysfunction) syndrome, other studies are necessary to better elucidate noni’s mechanisms of action, mainly due to the complexity of the pathophysiology of obesity and its metabolic dysfunction. In this review, we summarize not only the clinical effects, but also important cell signaling pathways in in vivo and in vitro assays of potent bioactive compounds present in the noni plant which have been reported in studies of obesity and obesity-associated metabolic dysfunction.

The activation of hepatic stellate cells (HSCs) is an important step in the progress of liver fibrosis. Fibrosis can be impeded by HSC reversion to a quiescent state or HSC clearance through apoptosis. To investigate the apoptotic effects of hsian‐tsao ( Mesona procumbens Hemsl) on human HSCs, the expression levels of cleaved caspase‐3, p38, and c‐Jun N‐terminal kinase (JNK) were assessed using Western blotting, and the caspase‐3 activity was measured using caspase‐3/CPP32 colorimetric assay kit. Hsian‐tsao extract (HTE) increased the activity of caspase‐3 and the level of activated caspase‐3, indicating the activation of apoptosis. The intracellular reactive oxygen species (ROS) level increased in a dose‐dependent manner. This increase was prevented by an antioxidant, suggesting that HTE induces ROS accumulation. In addition, we found that HTE induced the phosphorylation of the mitogen‐activated protein kinases JNK and p38. These collective data indicate that HTE induces apoptosis via ROS production through the p38, JNK, and caspase‐3‐dependent pathways. HTE may decrease HSC activation in liver fibrosis and may have a therapeutic potential.

This study identified and quantified five phenolic compounds, and evaluated the antioxidant capacity in vitro of fifteen native and exotic Brazilian fruit seeds that are typically discarded as waste. The contents of phenolic compounds were determined by ultra-performance liquid chromatography coupled with tandem mass spectrometry, and the antioxidant capacity was determined by oxygen radical absorbance capacity (ORAC) and 2,2-diphenyl-1-picrylhydrazyl (DPPH•) antioxidant assays. The results showed the antioxidant activity of Campomanesia pubescens, Hovenia dulcis Thunberg and Syzygium jambos (L.) Alston in the ORAC assay, and Hymenaea stigonocarpa, Hovenia dulcis Thunberg and Campomanesia pubescens in the DPPH• assay. Among the fifteen samples, four were highlighted regarding phenolic compound analyzes: Hovenia dulcis Thunberg (5.723 µg g-1) for gallic acid and myricetin (111.057 µg g-1), Passiflora edulis (1.208 µg g-1) for chlorogenic acid, Annona atemoya (1.0580 µg g-1) for vanillic acid, and Campomanesia pubescens (0.420 µg g-1) for ferulic acid. Therefore, these fruit seeds can be used as alternative sources of natural antioxidants.
