Anti‐Inflammatory and Anti‐Oxidant Effects of p‐Chloro‐phenyl‐selenoesterol on TNBS‐Induced Inflammatory Bowel Disease in Mice

Abstract: This study aims to investigate the protective effect of p-chloro-phenyl-selenoesterol [PCS; 0,2 mg/kg; 10 ml/kg i.g.) in colitis induced by 2,4,6-trinitrobenzene sulfonic acid [TNBS; 2 mg/100 µl 50% ethanol; intrarectally) in mice. Several parameters including weight, length, histological analyses determination, thiobarbituric acid reactive species, reactive species levels, superoxide dismutase, catalase, and myeloperoxidase (MPO) activity of colon were evaluated. The serum levels of tumor necrosis factor alph… Show more

“…74 Oxidative stress and its consequent lipid peroxidation are known to damage cellular macromolecules such as DNA, lipids, and proteins, which could exacerbate free radical chain reactions and activate the release of pro-inflammatory mediators, disrupt the integrity of the intestinal mucosal barrier, and activate inflammatory mediators. 75 MPO is an enzyme abundantly found in neutrophils which has been used as a marker of cell-specific infiltration and the severity of inflammation. 76 As a marker for free radicals-induced lipid peroxidation, MDA is widely used as a biochemical marker of oxidative stress.…”

“…35 In addition, the activation of these cells causes oxidative stress by increasing RS levels. 36 Research has suggested that antioxidant systems become impaired in RA, 33,37−40 and this is particularly important as our data revealed lower NPSH levels and SOD activity in the cerebral cortex, hippocampus, and paw tissues of CFAexposed. The results of the NPSH content analyzed after applying CFA and/or Se-DMC or meloxicam treatments are listed in Table 1.…”

“…There is a close link between oxidative stress and inflammation, and NF-κB plays a vital role in this process by balancing the synthesis and release of proinflammatory substances . In addition, the activation of these cells causes oxidative stress by increasing RS levels …”

“…Although some evaluated parameters did not differ significantly among the different treatments with L. lactis, it was noteworthy that compared with the C+ group, the NFXi group presented a decrease in the severity of inflammation as indicated by reduced body weight loss and decreases in macroscopic and histological scores. The evaluation of these parameters was important because colitis caused by intrarectal administration of TNBS causes abrupt weight loss, rectal prolapse and transmural inflammation (Kiesler et al 2015;Zarzecki et al 2017).…”
