Anti-Inflammatory Drugs Remodel the Tumor Immune Environment to Enhance Immune Checkpoint Blockade Efficacy

Abstract: Identifying strategies to improve the efficacy of immune checkpoint blockade (ICB) remains a major clinical need. Here, we show that therapeutically targeting the COX-2/PGE 2 /EP2-4 pathway with widely used non-steroidal and steroidal antiinflammatory drugs synergized with ICB in mouse cancer models. We exploited a bilateral surgery model to distinguish responders from non-responders shortly following treatment and identified acute IFN-γ-driven transcriptional remodeling in responder mice, which was also assoc… Show more

“…Driving the formation of “hot” tumors is a gradual and complicated process. Extensive efforts have been made to convert cold tumors to hot tumors, but the clinical applications are limited [ 25 ]. However, using reliable biomarkers to distinguish “hot” and “cold” tumors is a convenient and feasible strategy.…”

“…Cancer cell-intrinsic cyclooxygenases-2 expression contributed to resistance to immune checkpoint blockade ( 75 ). Therapeutically targeting the cyclooxygenases-2 pathway with widely used non-steroidal and steroidal anti-inflammatory drugs was able to synergize with immune checkpoint blockers and strengthen their anti-tumor effects in mouse CRC models ( 76 ). Celecoxib is a non-steroidal anti-inflammatory drug.…”

“…Many previously published literatures have indicated that the components of TME are dynamically changing and related to a variety of biological behaviors of cancers such as invasion, metastasis, and immune escape of immunological surveillance ( 17 , 18 ). So far, according to the status of tumor-infiltrating immune cells (TICs) in TME, tumors can be divided into two different types: hot tumors and cold tumors ( 19 , 20 ). Hot tumor possesses a high density of CD8 + T lymphocytes in tumor tissue whose function is weakened by immunosuppressive molecules.…”
