Anti-inflammatory effect of fullerene C60 in a mice model of atopic dermatitis

Abstract: Background Water-soluble form of fullerene C60 is a promising tool for the control of ROS-dependent inflammation including allergic diseases. Anti-inflammatory effects of C60 (nC60) aqueous dispersion were evaluated in the mouse models of atopic dermatitis using subcutaneous (SC) and epicutaneous (EC) applications during 50 days period. A highly stable nC60 was prepared by exhaustive dialysis of water-organic C60 solution against water, where the size and ζ-potential of fullerene nanoparticles are about 100 nm… Show more

“…In addition, an increase of IFN‐γ was observed in the splenocytes, whereas the production of IL‐12 was comparable. Consequently, restoring of Th2/Th1 balance in the groups treated with PBE was found through the serum IgG 2a /IgG 1 ratio, which is marker for Th1 and Th2 lymphocytes, respectively (Shershakova et al, ). In contrast, PD inhibited not only splenic production of Th2 cytokines and serum level of IgG 1 but also splenic production of IFN‐γ and serum level of IgG 2a .…”

“…Due to blockade of signaling molecules and cytoplasmic reactive oxygen species levels, fullerenes prevented the in vivo release of histamine and anaphylaxis. In two other studies, treatment of OVA‐sensitized mice with tetraglycolate fullerenes or a water‐soluble form of C60 fullerene resulted in a shift in the cytokine production from Th2 to Th1 as well as reduced airway inflammation, eosinophilia, and bronchoconstriction triggered by the production of an anti‐inflammatory P‐450 eicosanoid metabolite or an increase in Foxp3 and filaggrin mRNA, respectively …”

“…Two recent studies using the OVA asthma model with TiO 2 nanoparticles (primary particle size 22 nm) and graphene oxide (particle thickness 0.61 nm, other dimensions between 20 nm and 4 µm) showed similar results and a shift from a Th2‐dominated reaction to a Th1 pattern with fewer eosinophils but more neutrophils as a result of nanoparticle treatment . Also in other contexts, immunomodulation with Th1 shifts has been observed for nanoparticles of very different chemistry and size, such as sulfonate‐ and phosphonate‐functionalized polystyrene nanoparticles (245 and 227 nm), iron oxide nanoparticles (100 nm), aqueous fullerene C 60 nanoparticles (100 nm), and selenium nanoparticles (60 nm) . It is not clear whether the immunomodulation depends on specific nanoparticle properties, such as a negative surface charge as the only common feature of the above‐mentioned particles, or whether this is a more unspecific phenomenon.…”
