Background: Stunting causes growth and development issues which are characterized by a systemic inflammatory response and neurotransmitter disorder. The South Kalimantan natural resources i.e. extracts of Pasak Bumi (Eurycoma longifolia Jack) and seluang fish have the potential as a modified therapy for stunting management.Objective: To analyze the effect of Pasak Bumi and seluang fish administration in overcoming systemic inflammation and neurotransmitter disorders in the stunting rats model.Methods: Rats were divided into six group i.e Positive Control (KP): Stunting rats + placebo + standard feed; (P1): stunting rats + 70% EPB 15 mg/kg body weight (BW) + standard feed; (P2): Stunting rats + DHA 1 mg/kg BW + standard feed; (P3): Stunting rats + EPB 15 mg/kg BW + DHA 1 mg/kg BW + standard feed; (P4): Stunting rats + seluang fish; (P5): Stunting rats + EPB 15 mg/kg BW + seluang fish for 5 weeks; Parameters measured were levels of IL-6, TNF-α, and serotonin.Results: There were significant differences in levels of TNF-α (p = 0.000) and serotonin (p = 0.000).Conclusion: The administration of Pasak Bumi extract and seluang fish was able to inhibit systemic inflammation and neurotransmitter dysregulation.

Eurycoma longifolia (EL) and Eurycoma harmandiana (EH) are natural medicinal plants belonging to the Simaroubaceae family, and are well‐known for their ability to enhance male sexual performance. The present study investigated the phosphodiesterase‐5 (PDE‐5) inhibitory activity of intact roots of EL and EH. Additionally, canthin‐6‐one alkaloids, β‐carboline alkaloids, and quassinoids were also screened for PDE‐5 inhibitory activity. We developed in vitro root and callus cultures of EL and EH to determine their PDE‐5 inhibitory activity. Our results indicated that canthin‐6‐one alkaloids, which include canthin‐6‐one‐9‐O‐β‐D‐glucopyranoside, 9‐methoxycanthin‐6‐one, canthin‐6‐one, and 9‐hydroxycanthin‐6‐one, exhibited PDE‐5 enzymatic inhibitory activity, with IC50 values of 2.86±0.23, 3.30±1.03, 4.31±0.52, and 4.66±1.13 μM, respectively. The ethanolic extract of the intact roots of EL and EH, and the in vitro root culture of EH had large amounts of canthin‐6‐one alkaloids (1.50±0.04, 2.12±0.03, and 3.48±0.08 mg/g dry weight, respectively), and showed potent PDE‐5 inhibition. Our findings indicate that in vitro root cultures of EH may be used to replace intact plants, and canthin‐6‐one‐9‐O‐β‐D‐glucopyranoside should be further investigated for development as a health supplement.

Piper longum L.(PL) is considered one of the most important species traditionally used for treating various ailments and has indicated the presence of alkaloids, flavonoids, and steroids. In this study, we isolated the chemical compounds of PLleaves,andmeasuredNO, IL‐6, iNOS, as well as COX‐2 protein levels. In addition, molecular docking analysis were used to further understand anti‐inflammation effect of the compounds. We identified one new alkaloid named piperlongumine A (1) with ten known compounds (2‐11). The new compound (1) and two other alkaloids 2E)‐3‐(4‐hydroxy‐3‐methoxyphenyl)‐1‐(pyrrol‐1‐yl)propanone (7) and piperchabamide A (8) significantlyreduced NO production in LPS‐stimulated RAW 264.7 cells with the IC50 values of 0.97 ± 0.05 mM, 0.91 ± 0.07mM, 1.63 ± 0.14 mM, respectively. Moreover, at concentration of 2 mM, compound 1 inhibited approximately 98 ± 0.64 % of IL‐6 secretion, and decreased iNOS and COX‐2 protein level by about 96 and 19 folds compared to LPS treatment alone, respectively. Furthermore, compounds 1, 7, and 8 were predicted to bind and inhibit IL‐6, TNF‐a, and iNOS, with compound 1 showing the highest binding energy of ‐7.09 kcal/mol. This study provides new insights for potential anti‐inflammatory drug design and warrants further investigation.
