Tissue toxicity caused by 5-fluorouracil (5-FU) is associated with increased reactive oxygen species and inflammatory cytokines. Sinapic acid (SA) has both antioxidant and anti-inflammatory activities. Although SA has been shown to ameliorate chemical-induced tissue damage in various experimental models, its effects against 5-FU-induced ovarian damage have not yet been investigated. It was therefore aimed to evaluate the therapeutic potential of SA against 5-FU-induced ovarian damage in rats, together with the mechanisms of oxidative stress and inflammation for the first time in this study. Thirty rats were distributed into five groups: control, 5-FU (100 mg/kg) 5-FU+SA (2.5 and 5 mg/kg) and SA (5 mg/kg). 5-FU was applied to rats intraperitoneally on the 1st day of experiments and then SA was administrated for 3 successive days. Ovarian levels of lipid peroxidation [malondialdehyde (MDA)], oxidative stress (total oxidant status (TOS) and oxidative stress index (OSI)] antioxidant system [total antioxidant status (TAS), and catalase (CAT)], DNA damage [8-hydroxy-2'-deoxyguanosine (8-OHdG)] and inflammatory [interleukin-6 (IL-6)] markers in ovarian tissues were determined using spectrophotometric methods. To investigate the mechanism of the Aucklandiae Radix–Amomi Fructus (AR–AF) herb pair in treating gastric cancer (GC) by using network pharmacology and experimental verification. Using the traditional Chinese medicine system pharmacology database and analysis platform (TCMSP), the major active components and their corresponding targets were estimated and screened out. Using Cytoscape 3.7.2 software, a visual network was established using the active components of AR–AF and the targets of GC. Based on STRING online database, the protein interaction network of vital targets was built and analyzed. With the Database for Annotation, Visualization, and Integrated Discovery (DAVID) server, the gene ontology (GO) biological processes and the Kyoto Encyclopedia of Genes and Genomes (KEGG) signaling pathways of the target enrichment were performed. AutoDock Vina was used to perform molecular docking and calculate the binding affinity. The mRNA and protein expression levels of the hub targets were analyzed by the Oncomine, GEPIA, HPA databases and TIMER online tool, and the predicted targets were verified by qRT–PCR in vitro. Eremanthin, cynaropicrin, and aceteugenol were identified as vital active compounds, and AKT1, MAPK3, IL6, MAPK1, as well as EGFR were considered as the major targets. These targets exerted therapeutic effects on GC by regulating the cAMP signaling pathway, and PI3K-Akt signaling pathway. Molecular docking revealed that these active compounds and targets showed good binding interactions. The validation in different databases showed that most of the results were consistent with this paper. The experimental results confirmed that eremanthin could inhibit the proliferation of AGS by reducing the mRNA expression of hub targets. As predicted by network pharmacology and validated by the experimental results, AR–AF exerts antitumor effects through multiple components, targets, and pathways, thereby providing novel ideas and clues for the development of preparations and the treatment of GC.

Because of the complex etiology, the treatment of gastric cancer is a formidable challenge for contemporary medical. The current treatment method focuses on traditional surgical procedures, supplemented by other treatments. Among these other treatments, Traditional Chinese Medicine (TCM) plays an important role. Here, we used the systems pharmacology approach to reveal the potential molecular mechanism of PRGRC on gastric cancer which composes of Pinellia ternata(Thunb.) Breit., Rheum palmatumL., Gentiana scabraBunge, Radix Aucklandiae and Citrus aurantium L. This approach combines pharmacokinetics analysis with pharmacodynamics evaluation for the active compounds screening, targets prediction and pathways assessing. Firstly, through pharmacokinetic evaluation and target prediction models, 83 potential compounds and 184 gastric cancer-related targets were screened out. Then, the results of network analysis suggested that the targets of PRGRC were mainly involved two aspects: apoptosis and inflammation. Finally, we verified the reliability of the above analysis at the cellular level by using naringenin and luteolin with good pharmacokinetic activity as representative compounds. Overall, we found that PRGRC could influence the development of gastric cancer from a multi-scale perspective. This study provided a new direction for analyzing the mechanism of TCM.
