Piper aduncum L. is widely distributed in tropical regions and the ethnobotanical uses of this species encompass medicinal applications for the treatment of respiratory, antimicrobial, and gynecological diseases. Chemical studies reveal a diverse array of secondary metabolites, including terpenes, flavonoids, and prenylated compounds. Extracts from P. aduncum have shown antibacterial, antifungal, and larvicidal activities. Our study explores the activity of extracts and partitions against Mycobacterium tuberculosis H37Rv, as well as the chemical diversity of the bioactive partition. This marks the first investigation of the bioactive partition of P. aduncum from agroecological cultivation. The ethyl acetate partition from the ethanolic leaf extract (PAEPL) was found to be the most active. PAEPL was subjected to column chromatography using Sephadex LH-20 and the obtained fractions were analyzed using UHPLC-HRMS/MS. The MS/MS data from the fractions were submitted to the online GNPS platform for the generation of the molecular network, which displayed 1714 nodes and 167 clusters. Compounds were identified via manual inspection and different libraries, allowing the annotation of 83 compounds, including flavonoids, benzoic acid derivatives, glycosides, free fatty acids, and glycerol-esterified fatty acids. This study provides the first chemical fingerprint of an antimycobacterial sample from P. aduncum cultivated in an agroecological system.

The present study thus aimed at the development and physicochemical characterization of solid lipid nanoparticles loaded with crude extract of Piper corcovadensisroots (SLN-CEPc) and chitosan-coated solid lipid nanoparticles loaded withcrude extract of P. corcovadensisroots (C-SLN-CEPc), as well as the determination of its antimycobacterial activity against Mycobacterium tuberculosisH37Rv, its cytotoxicity against the Vero cell line and evaluation in the hemolysis assay. Both formulations containing the encapsulated extract showed high encapsulation efficiency, formed by a monodispersed system with small and spherical particles, and there was no aggregation of particles. In the biological assays, SLN-CEPc and C-SLN-CEPc showed promising anti-M. tuberculosisactivity with a minimum inhibitory concentration (MIC) of 12.5 μg/mL, whereas the cytotoxic concentrations obtained at 50% (CC50) in Vero cells were 60.0 and 70.0 μg/mL, respectively. Therefore, nanoencapsulation showed satisfactory results, justifying its usage in the development of new products.

Piper multinodum C.DC. belongs to the Piperaceae family, and is known as "Jaborandi-manso". This species has no reported botanical, chemical, pharmacological and/or toxicological scientific studies, and has restricted occurrence in degraded Brazilian biomes. The Piper genus is known to be aromatic, the essential oils (EO) obtained from different organs have demonstrated significant biological activities, and can be an important tool for chemophenetic and ecological definitions. The present study aimed to characterize the chemical profile of the EO from different parts of P. multinodum and their activity against Mycobacterium tuberculosis H 37 Rv (ATCC, 25618). The EO were obtained by hydrodistillation and characterized by GC-MS and GC-FID. Chemical composition of the volatile mixture showed a great diversity of compounds that diverged between the vegetative and reproductive parts. The α-pinene compound was identified as being the most predominant in the leaves, infructescences and inflorescences (32.49, 67.23 and 40.23%, respectively). The branches (secondary stem) showed to be rich in sesquiterpenes and monoterpenes: α-copaene (13.24%), E-caryophyllene (12.32%), α-pinene (20.34%), and myrcene (11.23%). The chemical profile of stems and roots showed a low percentage of monoterpenes and sesquiterpenes, but a high percentage of arylpropanoids, with E-methyl-isoeugenol (77.58%) registered in the stems and euasarone (81.34%) in the root. The antimycobacterial activity showed the highest activities recorded for the EO from roots (78.51 µg/mL) and infructescences (85.91 µg/mL). In addition to the findings related to biological activity, the determination of chemical diversification between the different parts of P. multinodum may help to understand the ecological issues of the Piperaceae family as well as to improve the chemotaxonomic knowledge of the genus Piper.
