Anti-senescence role of heterozygous fumarate hydratase gene knockout in rat lung fibroblasts in vitro

Abstract: Abnormalities in tricarboxylic acid (TCA) cycle function were related to a variety of pathological processes. Fumarate hydratase (FH) is a required enzyme in the TCA cycle. To explore the general influence of FH knockout, we isolated FH+/– rat and normal rat lung fibroblasts and cultured these cells in vitro. The isolated fibroblasts with the current method were rather homogeneous and were confirmed spindle in morphology, positive for vimentin and negative for α-SMA (α-smooth muscle actin). Sequencing of the P… Show more

“…Studies have reported that PTX3 can be produced by innate immune cells and plays an important role in innate immunity, regulation of inflammation, and matrix deposition through the interaction with several ligands. 7 PTX3 contributes to tumor-related inflammation, and its expression has been identified as a new diagnostic and prognostic biomarker for various types of cancer, including glioma, 11 prostate cancer, 12 lung cancer, 13 soft-tissue liposarcoma, 14 and pancreatic cancer. 15 Recent evidence reveals that the expression of PTX3 in tumors and adjacent liver tissues of patients with hepatocellular carcinoma is significantly up-regulated.…”

“…Fumarate hydratase (FH) is a key enzyme in the TCA cycle, which can convert fumarate into malic acid. FH +/- rat primary fibroblasts exhibited anti-aging properties [ 41 ]. Pyruvate produced by glycolysis can be converted into citrate after entering the mitochondria, which participates in Krebs through IDH3.…”

“…Thus, we suggest that CD38 decreased NAD+ levels and Sirt6 expression to stimulate cell senescence. Other researchers also detected the expression of CD38 in H9C2 cells [ 48 ]. Some studies reported that FoxO3a, a transcription factor, bound to the promoter region of Sirt6 and enhanced its expression [ 49 , 50 ].…”
