Anti-tumor Efficiency of Lipid-coated Cisplatin Nanoparticles Co-loaded with MicroRNA-375

Abstract: One of the major challenges in the hepatocellular carcinoma (HCC) treatment is its insensitivity to chemotherapeutic drugs. Here, we report the development of novel lipid-coated cisplatin nanoparticles co-loaded with microRNA-375 (NPC/miR-375) as a potential treatment for chemotherapy insensitive HCC. The NPC/miR-375 was fabricated by mixing two reverse microemulsions containing KCl solution and a highly soluble cis-diaminedihydroplatinum (II) coated with a cationic lipid layer. Subsequently, the miR-375 was i… Show more

“…Overexpression of miR-375 has been shown to inhibit AEG-1 and MDR1 expression in HCC cells. 18,22 As miR-375 is an endogenous miRNA that is downregulated in HCC, restoration of miR-375 may serve as a credible strategy to overcome MDR.…”

“…In addition, previous work has suggested a pro-cell death role of this microRNA. For example, Yang and colleagues (28) showed very recently that miR-375 could enhance the anti-tumor effect of cisplatin by inhibiting cell proliferation and inducing apoptosis and cell cycle arrest in hepatocellular carcinoma. However, the role of miR-375 in other tissues or cells was unknown.…”

“…For instance, survivin inhibitor YM155 and 3′,4′,5′,5,7-pentamethoxyflavone were shown to have the ability to sensitize cancer cells to CDDP. 18,40 Previously, our group has also improved CDDP efficacy in hypoxia by the combined use of metformin to inhibit the activation of NF-κB/HIF-1α signaling. 12 However, the systemic toxicity of CDDP cannot be avoided.…”
