Background and Purpose: Candida species constitute an important group of opportunistic fungi, which cause various clinical diseases. Considering the resistance of some Candida species to conventional antifungal agents, treatment of such cases may be challenging and complicated. The purpose of this study was to evaluate and compare the antifungal activities of Euphorbia macroclada latex and fluconazole against different Candida species.Materials and Methods:A total of 150 Candida isolates including C. albicans (n=77), C. glabrata (n=28), C. parapsilosis (n=23), C. tropicalis (n=15), C. krusei (n=4), C. famata (n=1), C. kefyr (n=1) and C. inconspicua (n=1) were included in this study. In vitro antifungal activities of Euphorbia macroclada latex and fluconazole against these Candida species were evaluated, according to M27-A2 protocol on broth macrodilution method by the Clinical and Laboratory Standards Institute (CLSI). Results:Among 150 Candida isolates, 98 isolates (65.33%), i.e., C. albicans (n=41), C. glabrata (n=23), C. tropicalis (n=12) and C. parapsilosis (n=22) with minimal inhibitory concentration (MIC) ≤ 8 μg/ml were susceptible to fluconazole. Resistance to fluconazole was noted in 15 isolates, i.e., C. albicans (n=10), C. glabrata (n=2), C. krusei (n=1), C. kefyr (n=1), and C. inconspicua (n=1), with MICs of 64 µg/ml. The remaining isolates (n=37) including C. albicans (n=26), C. glabrata (n=3), C. tropicalis (n=3), C. parapsilosis (n=1), C. krusei (n=3) and C. famata (n=1) with MIC= 16-32 µg/ml showed dose-dependent susceptibility. The latex of Euphorbia macroclada was able to inhibit the growth of 30 out of 150 tested Candida isolates with MIC range of 128-512 µg/ml. These isolates were as follows: C. albicans (n=2), C. glabrata (n=4), C. parapsilosis (n=19), C. krusei (n=2) and C. tropicalis (n=3). Compared to other isolates, higher MIC values were noted for C. albicans and C. glabrata (512 µg/ml), respectively.Conclusion:The latex of Euphorbia macroclada showed notable antifungal activities against some pathogenic Candida species. Therefore, it can be potentially used as an alternative antifungal agent in future. However, further research is required to identify its active components.

Background/Aim:To determine the anti-hyperglycemic effect of Euphorbia antiquorum L. root.Materials and Methods:The study evaluates the anti-hyperglycemic effect of E. antiquorum root in streptozotocin-nicotinamide-induced Type 2 diabetes mellitus and fructose-induced insulin resistance models. Alcohol and aqueous extracts of E. antiquorum root were administered at doses 200 and 400 mg/kg p.o. Serum levels of glucose, total cholesterol, triglycerides, glycosylated hemoglobin (GHb), and hepatic levels of malondialdehyde, glutathione, and glycogen were estimated.Results:Treatment with the alcohol and aqueous extracts of E. antiquorum roots resulted in significant (P < 0.001) lowering of serum blood glucose and GHb levels in both the models. Flavonoids, phenolic compounds, and glycosides were detected in the preliminary phytochemical screening.Conclusion:Root of E. antiquorum showed promising anti-hyperglycemic effect which may be due to the presence of important phytochemicals.

The emergence of multidrug resistant bacterial pathogens and the increase of antimicrobial resistance constitutes a major health challenge, leading to intense research efforts being focused on the discovery of novel antimicrobial compounds. In this study, endophytes were isolated from different parts of Sandwithia guyanensis plant (leaves, wood and latex) belonging to the Euphorbiaceae family and known to produce antimicrobial compounds, and chemically characterised using Molecular Network in order to discover novel antimicrobial molecules. One fungal endophyte extract obtained from S. guyanensis latex showed significant antimicrobial activity with Minimal Inhibitory Concentration on methicillin-resistant Staphylococcus aureus at 16 µg/mL. The chemical investigation of this fungus (Lecanicillium genus) extract led to the isolation of 5 stephensiolides compounds, four of which demonstrated antibacterial activity. Stephensiolide I and G showed the highest antibacterial activity on MRSA with a MIC at 4 and 16 µg/mL respectively.Plants 2020, 9, 47 2 of 11 chemical constituents of a wide variety of extracts in a single map [5,6]. Furthermore, MN offers the possibility to map additional information, such as biological, analytical and taxonomical details over networks [6][7][8]. In this study, we applied a related multi-informative prioritisation strategy based on the use of MN, with the aim of finding new antimicrobial structures from of a set of extracts from 42 Euphorbiaceae endophytes.The Euphorbiaceae family comprises approximately 283 genera and 7300 species, and has been found in various morphological types, including herbs, shrubs and trees [9]. Some species of the family are used as lubricants and purgatives, while others are used as a source of medicinal drugs for the treatment of diarrhoea, inflammation and swelling. Their milky latex secretions impart several toxic effects such as dermatitis, severe irritation to the eyes and stomach discomfort. Moreover, Euphorbia sp. latex has been shown to possess moderate antimicrobial properties and ability to protect against herbivores [10]. Together, the antimicrobial, insecticidal and toxicity properties of Euphorbia latex makes it a hostile environment for microbes and plant-feeding insect pests. However, it has been recently highlighted that Euphorbia sp. latex contains unexpectedly complex bacterial and fungal communities [11]. Meanwhile, endophytes, which are microorganisms residing in plant tissues without inducing any diseases in the host plants, have been shown to defend themselves as well the plants from pathogens and have a crucial role in plant survival. They also improve the ecological adaptability of the hosts by enhancing their tolerance to environmental stresses and resistance to phytopathogens and/or herbivores [12].
