Streptomyces are well known for their competence to produce thousands of bioactive secondary metabolites and enzymes. This study aimed to assess the inhibitory activities of crude extracts from diverse Streptomyces collected from rice soils in Narathiwat, Thailand, against foodborne bacterial pathogens. In total, 136 Actinomycete isolates were screened using a cross-streak method for the ability to produce effective metabolites against 5 pathogenic bacteria. Out of these, 19 (13.97%) isolates had antibacterial activity against at least one tested bacterium. Most of the isolates could strongly suppress the growth of S. aureus ATCC25923 and B. cereus MTCC430 except P. aeruginosa ATCC27853. On the basis of morphological, cultural, and biochemical characteristics, all potent isolates exhibited typical features that fitted the genus Streptomyces. Two of the 7 selected ethyl acetate crude extracts had good antagonistic activity against S. aureus ATCC25923 and B. cereus MTCC430 when tested using the agar well diffusion assay. Furthermore, minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values of the 2 extracts evaluated using the colorimetric broth microdilution method ranged from 256 to >1,024 μg/ml against the tested bacteria. The partial nucleotide sequences of the 16S rRNA gene led to identifying both active isolates as Streptomyces species. These active Streptomyces isolates could provide an interesting source for generating innumerable natural compounds with antibacterial activity that can presumably be developed to fight bacterial pathogens in the near future.

Antibiotic-resistant strains are a global health-threatening problem. Drug-resistant microbes have compromised the control of infectious diseases. Therefore, the search for a novel class of antibiotic drugs is necessary. Streptomycetes have been described as the richest source of bioactive compounds, including antibiotics. This study was aimed to characterize the antibacterial compounds of Streptomyces sp. PJ85 isolated from dry dipterocarp forest soil in Northeast Thailand. The 16S rRNA gene sequence and phylogenetic analysis showed that PJ85 possessed a high similarity to Streptomyces actinomycinicus RCU-197T of 98.90%. The PJ85 strain was shown to produce antibacterial compounds that were active against Gram-positive bacteria including methicillin-resistant Staphylococcus aureus (MRSA). The active compounds of PJ85 were extracted and purified using silica gel column chromatography. Two active antibacterial compounds, compound 1 and compound PJ85_F39, were purified and characterized with spectroscopy, including liquid chromatography and mass spectrometry (LC–MS). Compound 1 was identified as actinomycin D, and compound PJ85_F39 was identified as dihomo-γ-linolenic acid (DGLA). To the best of our knowledge, this is the first report of the purification and characterization of the antibacterial compounds of S. actinomycinicus.

Background: Natural pigments from microbial sources are a class of compounds with various structures and a wide range of uses, which have become an important source of antibiotic production. Objectives: This study aimed to screen and identify a new type of pigment-producing microorganism from purple soil in the Sichuan Basin. At the same time, this study aimed to purify and identify the produced pigment, preliminarily optimize its fermentation conditions, and characterize its antibacterial activity. Methods: Through morphology, physiology, biochemistry, and molecular sequencing, the strain LS-2 was identified. The pigment was purified by column chromatography, and its antibacterial activity was characterized by a disc diffusion assay. Through the analysis of the ultraviolet-visible (UV-vis) absorption spectrum, infrared (IR) absorption spectrum, and liquid chromatography-mass spectrometry (LC-MS), the yellow pigment was identified. Results: An actinomycin D-producing Streptomyces parvulus was isolated from the purple soil in the Sichuan Basin. Its optimal fermentation conditions were culture at 34°C on Gause’s synthetic medium with pH 6.5, and the yield reached 540 mg/L after 48 hours. The actinomycin D produced by the strain has excellent antibacterial activity against gram-positive (Staphylococcus aureus) and gram-negative (Escherichia coli). Conclusions: A new actinomycin D-producing strain of Streptomyces parvulus LS-2 was isolated and identified. The strain has a high production of actinomycin D, which has excellent antibacterial activity.
