Antibodies mediate intracellular immunity through tripartite motif-containing 21 (TRIM21)

Abstract: Antibodies provide effective antiviral immunity despite the fact that viruses escape into cells when they infect. Here we show that antibodies remain attached to viruses after cell infection and mediate an intracellular immune response that disables virions in the cytosol. We have discovered that cells possess a cytosolic IgG receptor, tripartite motif-containing 21 (TRIM21), which binds to antibodies with a higher affinity than any other IgG receptor in the human body. TRIM21 rapidly recruits to incoming anti… Show more

“…Our results would suggest that other UNG-deficient patients might have higher levels of some natural Ig classes. These switched Ig but also IgM could undergo normal affinity maturation and contribute to immunity by traditional means, but also by engaging intracellular receptors (52), which might be sufficient to make these patients appear fairly normal in the modern sanitary conditions. Finally, we find that Ung 2/2 mice have circulating autoantibodies to the same extent as Aicda 2/2 mice.…”

“…Furthermore, anti-DNA antibodies could enter nucleus via caveoli-mediated process and subsequently modulate expression of caveolin and p53 [11]. Recently, tripartite motif-containing 21 (TRIM21) was shown to be able to mediate antibody function inside cells by engaging with a potent effecter mechanism that targets the virus for degradation [12].…”
