Antibodies reacting with Simian Virus 40 capsid protein mimotopes in serum samples from patients affected by uveal melanoma

Abstract: The uveal melanoma (UM) is the most common human intraocular tumour. Simian Virus 40 (SV-40) is a small DNA tumor virus detected in some malignancies, including the cutaneous melanoma. In this study an indirect ELISA using synthetic peptides that mimic SV-40 antigens, was employed to detect antibodies against SV-40 in serum samples from UM patients. Our report indicates a significant higher prevalence of antibodies against SV-40 capsid protein antigens in serum samples from UM patients compared to controls. Ou… Show more

“…In this context, it should be noted that in a previous investigation we did not find any association between CN and other small DNA tumor viruses, such as the polyomavirus SV40. Indeed, the prevalence of SV40 antibodies in serum samples from CN patients (17%) did not differ from that revealed in HS (15%; Bononi et al, 2014 ).…”

“…To better elucidate these controversies, novel indirect ELISA tests employing synthetic peptides as mimotopes/specific SV40 antigens were set up (191, 192). These immunological and specific assays established that anti-SV40 antibodies can be revealed in human sera from patients affected by different tumors (211, 218, 223, 225, 237, 246, 252, 253, 280, 281), of the same kind found to be SV40-positive by molecular biology techniques. A significantly higher SV40 antibody prevalence was detected in sera from MPM, glioblastoma multiforme, osteosarcoma, and non-Hodgkin's lymphoma patients compared to age-/gender-matched healthy subjects (218, 225, 237, 252, 253, 280).…”

“…Plasma samples were prepared as previously described with modifications. 51 , 52 , 53 The supernatant was used for targeted metabolic profiling of 140 lipids and amino acids with an Acquity ultra performance liquid chromatography coupled to a Xevo TQ-S mass spectrometer (UPLC-TQ-MS, Waters Corp., Milford, MA, USA). 53 Data generated by UPLC-TQ-MS were quantitative data and are expressed in μM.…”
