Antibodies to Merkel Cell Polyomavirus Correlate to Presence of Viral DNA in the Skin

Abstract: To validate whether Merkel cell polyomavirus (MCV) serology correlates with MCV infection, we compared real-time polymerase chain reaction results for MCV DNA on fresh-frozen biopsy specimens from various skin lesions and healthy skin from 434 patients to MCV serology results using viruslike particles (VLPs) and MCV neutralization assays. Sixty-five percent of participants were MCV seropositive and 18% were MCV DNA positive. The presence of antibodies was correlated with the presence of virus DNA (odds ratio, … Show more

“…The seroprevalence for MCV among children was around 30 %, which is also in agreement with other studies (23). Seroprevalences of JCV and MCV in the adult Slovenian women were also similar (around 65 %) to other studies of European adults (24).…”

“…A correlation between MCPyV loads and antibody titers has been reported (22,23), suggesting that life-long persistence of MCPyV infection is correlated with higher antibody levels. The increase in MCPyV antibody levels with age may thus reflect the fact that subjects with high antibody titers had active viral replication associated with VP1 production, a state more frequently observed in older subjects due to waning immunity.…”

“…MCPyV DNA has been detected at varying levels of frequency (0-100%) in skin samples, and in anal and penile swabs (1,9,20,21,26,(32)(33)(34)(35)(36)(37)(38). The MCPyV viral load on the skin surface varies from less than 1 copy per 1000 cells to 1000 copies per cell (9,26,30,(39)(40)(41).…”
