Antibodies to the Glycoprotein GP2 Subunit Cross-React between Old and New World Arenaviruses

Abstract: Several viruses in the Arenaviridae family infect humans and cause severe hemorrhagic fevers which lead to high case fatality rates. Due to their pathogenicity and geographic tropisms, these viruses remain very understudied. As a result, an effective vaccine or therapy is urgently needed. Here, we describe efforts to produce cross-reactive monoclonal antibodies that bind to both New and Old World arenaviruses. All of our MAbs seem to be nonneutralizing and nonprotective and target subunit 2 of the glycoprotein… Show more

“…Recombinant proteins were produced using the well-established baculovirus expression system and this system has been published in detail in refs. [20][21][22] , including a video guide. Recombinant proteins were also produced in Expi293F cells (Thermo Fisher Scientific) by transfections of these cells with purified DNA using an ExpiFectamine 293 Transfection Kit (Thermo Fisher Scientific).…”

“…The GP is also regarded as the primary immunogen for LASV vaccine development [36] because vaccines expressing LASV GP conferred a higher level of protection than those expressing LASV NP only [23,47]. GP1 is the most variable subunit, while the GP2 is the most conserved subunit among mammarenaviruses [14,54,55]. Synonymous single nucleotide variations (SNV) were found in similar rates in both subunits of the GP and most non-synonymous SNVs were found in the GP1 subunit among intra-host LASV strains [12].…”

“…However, reported nAbs rarely possess crossneutralization against them. MAbs targeting sequence-conserved GP2 are considered very promising, while antibodies that cross-react with both NW and Old World arenaviruses neutralize neither (Amanat et al, 2018). NAbs for JUNV simply targeting GP1 have not been reported to have cross-neutralization capabilities against the five arenaviruses except for one elicited by Candid#1 that neutralizes both JUNV and MACV with varying efficiency in vitro (Clark et al, 2018).…”
