Antibody response to cytomegalovirus polypeptides captured by monoclonal antibodies on the solid phase in enzyme immunoassays

Abstract: Antibodies to different cytomegalovirus (CMV) polypeptide antigens, captured by monoclonal antibodies coated on the solid phase of an enzyme immunoassay test, were analyzed in 42 serum pairs submitted for serodiagnosis of CMV infection. Three CMV antigens, captured on the solid phase by three monoclonal antibodies of different specificities, designated CH92-1, CH65-1, and CH16-1, were glycoproteins A (gA), gC, and gD, respectively; and one antigen, captured by CH23, was a polypeptide with an apparent molecular… Show more

“…Among them p150 was recognized by IgG present in the zero-time serum samples of all the patients with no correlation with the antibody levels detected by conventional serological procedures. The data obtained in the present investigation define that p150 is a good antigen in vivo as already suggested (3,9), and induces a long-lasting antibody response. Therefore, the presence of antibody against this polypeptide could be a very useful tool to assess the occurrence of a previous CMV infection.…”

“…Antibodies specific for all four recombinant proteins could be detected in only two patients, and two patients produced an-VOL. 2,1995 HUMORAL IMMUNE RESPONSES TO HCMV 215 tibodies to p52 alone. The highest IgM antibody levels found appeared to be directed against p52 and pp65.…”

“…However, one patient who had a VOL. 2,1995 HUMORAL IMMUNE RESPONSES TO HCMV 217 maximum antigenemia test of 4 positive cells per 50,000 leukocytes, which is very low for a primary CMV infection, had an early antibody response as tested in the recombinant proteinbased ELISA, predominantly to pp65 and IE2, both IgM and IgG classes. An interesting finding was that in patients with secondary CMV infections, the level of antibodies to linear epitopes was inversely correlated to the level of antigenemia but the level of antibodies to conformational epitopes was not.…”

“…HCMV glycoprotein B (gB) is a major component of the viral envelope that plays a central role in viral entry and in virus spread from cell-to-cell . In addition, gB is highly immunogenic in humans [Cremer et al, 1985;Gonczol et al, 1986Gonczol et al, , 1990Kniess et al, 1991;Navarro et al, 1997;Pereira et al, 1991] and elicits a dominant antiviral neutralizing response [Britt et al, 1988[Britt et al, , 1990Marshall et al, 1992Marshall et al, , 1994Rasmussen et al, 1991]. Consequently, specific anti-gB antibodies are likely to be involved in protection against HCMV.…”
