Anticancer effect of aloe-emodin on cervical cancer cells involves G2/M arrest and induction of differentiation

Abstract: Aim: The aim of this study was to investigate the effects of aloe-emodin, a natural compound from the root and rhizome of Rheum palmatum, on the growth of human cervical cancer cells, HeLa. Methods: HeLa cells were treated with various concentrations of aloe-emodin for 1-5 d, and cell growth was measured by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide assay. The long-term growth effect was investigated by crystal violet assay. The distributions of the cell cycle and apoptosis were analyzed b… Show more

“…Aloe-emodin induced G2/M arrest and differentiation of cervical cancer cells (52), while induced apoptosis of retina ganglion cells in glaucomatous patients through regulation of ERK phosphorylation (53). Aloe-emodin induced apoptosis, autophagy and differentiation of glioma cells by inhibiting the action of ERK (54).…”

“…Control cells were treated with solvent alone for the durations indicated above. Cell cycle was analyzed as previously described [20] . The cells were trypsinized, washed twice with ice cold PBS, fixed in 70% ethanol and stained with propidium iodide (PI; 5 μg/mL PI in PBS containing 0.1% Triton X-100 and 0.2 mg/mL RNase A) in the dark for 30 min at 4 °C.…”

“…Emodin is regarded as the most active constituent in Giant Knotweed Rhizome and exerts many potent biological effects [9,10] , such as anticancer [11] , antimicrobial [12] , and anti-inflammatory effects [13] . Several studies have revealed that emodin is efficacious in the management of hepatic fibrosis [14,15] .…”
