Anticancer effect of HOTTIP regulates human pancreatic cancer via the metabotropic glutamate receptor 1 pathway

Abstract: Abstract. The present study aimed to determine how the expression and function of HOTTIP modifies, and regulates the metabotropic glutamate receptor 1 (mGluR1) to affect human pancreatic cancer cell viability. HOTTIP expression was higher in human pancreatic cancer tissue compared with in para-carcinoma tissue. However, downregulation of HOTTIP expression was revealed to significantly reduce cell viability, induce apoptosis, promote caspase-3 and caspase-8 activities and increase Bax expression in pancreatic c… Show more

“…HOTTIP is another HOX-dependent lncRNA that is transcribed from the 5' end of the HOXA locus. HOTTIP controls the expression of several genes by interaction with polycomb repressive complex 2 (PRC2) and mixed lineage leukemia protein-1 (MLL1)/WD repeat domain 5 (WDR5) in this locus (36,37). This lncRNA is significantly expressed in pancreatic cancer tissues and cell lines compared to normal (37).…”

“…mGluR1 expression was detected in pancreatic cancer and shown to be regulated by the long non-coding RNA (lncRNA) HOXA distal transcript antisense RNA (HOTTIP) [ 133 , 134 , 135 ]. lncRNAs are byproducts of RNA polymerase II transcription, which are now understood to be important for transcriptional regulation, and this has been shown to be associated with the development of cancer [ 133 , 134 , 135 ]. In pancreatic cancer, the connection between mGluR1 and HOTTIP was suggested to be a potential prognostic marker [ 133 , 136 ].…”

In pancreatic cancer, the connection between mGluR1 and HOTTIP was suggested to be a potential prognostic marker [ 133 , 136 ]. Altered levels of HOTTIP are correlated with changes in mGluR1 expression, and these changes were linked to tumor cell viability, survival, migration, invasion, and apoptosis [ 133 , 136 ].…”

“…The downregulation of HOTTIP-induced apoptosis was confirmed by enhanced caspase-3/8 activity and Bax family protein expression in PC cells. In addition, mGluR1 levels also decrease with inactivation of the phosphoinositide 3-kinase (PI3K)/AKT/mTOR pathway and may also contribute to PC cell apoptosis [56]. In addition, there is evidence showing that the functional rs1859168 A>C polymorphism may decrease the risk of PC genesis by downregulating HOTTIP expression [57].…”
