Anticancer effects of myricetin derivatives in non‐small cell lung cancer in vitro and in vivo

Abstract: Lung cancer is the most common cause of cancer‐related deaths. Moreover, exploring efficient tumor‐killing drugs is urgently needed. In our study, several derivative compounds of myricetin were synthesized and tested. Experiments on non‐small cell lung cancer (NSCLC) showed that S4‐2‐2 (5,7‐dimethoxy‐3‐(4‐(methyl(1‐(naphthalen‐2‐ylsulfonyl)piperidin‐4‐yl)amino)butoxy)‐2‐(3,4,5‐trimethoxyphenyl)‐4H‐chromen‐4‐one) had the strongest effect on A549 cell inhibition across all compounds. Furthermore, S4‐2‐2‐treated … Show more

“…To further validate the therapeutic effects of myricetin in patient samples, we attempted to generate CML patient-derived xenograft mouse model but was not successful. Nevertheless, our in vivo findings are supported by the recent studies demonstrating the potent anticancer efficacy in ovarian cancer and non-small lung cancer in mice without causing toxicity [23,24].…”

“…Besides, number of apoptotic cells enhanced after S4-2-2 treatment. Furthermore, invasiveness of A549 cells was prevented and number of migrating cells was decreased after S4-2-2 treatment [102].…”

“…In line with the proapoptotic effect exhibited by myricetin on many tumor cell lines [ 62 , 68 , 69 , 70 , 71 , 72 , 73 , 74 , 75 , 76 , 77 ], this compound has also induced apoptosis in endothelial cells through a reactive oxygen species (ROS)-dependent mechanism [ 67 ]. Despite the fact that in vivo studies in rodents support the chemopreventive role of myricetin in different cancer models [ 78 , 79 , 80 , 81 , 82 , 83 , 84 , 85 ], there is no current clinical evidence of its preventive activity in cancer, nor are there any active clinical trials using this compound. Additional in vitro studies suggest a synergistic effect of myricetin with traditional anticancer drugs [ 86 ].…”
